Peixe-zebra é um modelo animal popular para a pesquisa sobre o desenvolvimento do olho por causa de sua rápida<em&gt; Ex utero</em&gt; Desenvolvimento e fecundidade bom. Por 3 dias após a fertilização (DPF), as larvas irá mostrar a primeira resposta visual. Muitos genes foram identificados até o controle de um desenvolvimento adequada para os olhos, mas estamos longe de um entendimento completo da arquitetura genética subjacente. Todo o genoma perfil de expressão gênica é uma ferramenta útil para elucidar rede regulatória genética para o desenvolvimento do olho. No entanto, o pequeno tamanho do olho embrionárias em peixes-zebra torna difícil obter tecidos oculares intacta e pura para análise de expressão. Por exemplo, o comprimento ântero-posterior do olho entre o dia 2 e 3 é apenas cerca de 200-300 mM, enquanto que o diâmetro da lente é mM menos 100. Todos os procedimentos podem ser feitos com uma pinça fina e material de laboratório em geral sob oculares standard. Para dissecção da retina, o RPE single-layer é removida e descascada por escovar a ação ea adesão preferencial dos remanescentes RPE para a superfície da placa de cultura para dissecção. Para RPE ligado à dissecção da retina, a adesão de RPE para a placa de dissecção é removido antes da dissecção de modo que a RPE pode ser completamente preservada com a retina. Uma ação de levantamento cuidadoso deste tecido pode eficientemente separar a coróide presuntivo e esclera. A lente pode ser removido em ambos os casos por uma agulha de tungstênio gravado quimicamente. Em resumo, nossa abordagem pode obter tecidos oculares intactos e tem sido utilizada com sucesso para estudar perfis de tecidos específicos expressão da retina zebrafish<sup&gt; 1, 2</sup&gt; E epitélio pigmentar da retina<sup&gt; 3</sup&gt;.
