live cell response to mechanical stimulation studied by integrated optical and atomic force microscopy

Om te begrijpen het mechanisme waarmee levende cellen zin mechanische krachten, en hoe ze reageren en zich aanpassen aan hun omgeving, een nieuwe technologie in staat om cellen gedrag te onderzoeken op sub-cellulair niveau met een hoge ruimtelijke en temporele resolutie is ontwikkeld. Zo werd een atomic force microscoop (AFM) geïntegreerd met een totale interne reflectie fluorescentie (TIRF) microscopie en snel draaiende schijf (FSD) confocale microscopie. Het geïntegreerde systeem is breed toepasbaar in een breed scala van moleculaire dynamische studies in alle aanhangend levende cellen, waardoor de directe optische beeldvorming van de cel responsen aan mechanische stimulatie in real-time. Belangrijke herschikking van de actine filamenten en focale adhesies bleek te wijten aan de lokale mechanische stimulatie bij de apicale celoppervlak dat veranderingen veroorzaakt in de cellulaire structuur in de cel lichaam. Deze innovatieve technieken zal nieuwe informatie voor inzicht in levende cellen herstructurering en dynamiek in reactie op mechanische kracht. Een gedetailleerd protocol en een representatieve dataset te tonen dat levende cellen als reactie op mechanische stimulatie worden gepresenteerd.
