in situ protocol for butterfly pupal wings using riboprobes

Hier presenteren wij, in video-formaat, een protocol voor het in situ hybridisaties in popstadium vleugels van de vlinder Bicyclus anynana met behulp van riboprobes. In situ hybridisaties, een steunpilaar van de ontwikkelingsbiologie, zijn nuttig om de ruimtelijke en temporele patronen van genexpressie studie in de ontwikkeling van weefsels op het niveau van transcriptie. Als antilichamen die het eiwit producten van gentranscriptie doelgroep zijn nog niet ontwikkeld en / of er meerdere genen exemplaren van een bepaald eiwit in het genoom die niet kan worden gedifferentieerd gebruik van beschikbare antilichamen, kan in situs plaats daarvan worden gebruikt. Terwijl de een in-situ techniek voor de larven vleugel disks beschikbaar is geweest voor de vlinder community voor meerdere jaren, heeft het huidige protocol is geoptimaliseerd voor de grotere en meer kwetsbare popstadium vleugels.

Om genexpressie te onderzoeken in het popstadium vleugel weefsel van Bicyclus anynana, presenteren we een geoptimaliseerd protocol voor in-situ hybridisaties met behulp van riboprobes. We bieden ook richtlijnen voor de verdere optimalisatie van dit protocol voor het gebruik in popstadium vleugels van andere lepidoptera soorten.
