De meest kritische fase van de procedure is de generatie van verse GST-RBD. Gebrek aan aandacht te besteden aan deze stap is de belangrijkste reden voor de mislukking. rhoc gtpase activation assay

RhoC GTPase heeft 91% homologie met RhoA GTPase. Omwille van de prevalentie in cellen, hebben veel reagentia en technieken voor het RhoA GTPase ontwikkeld. Echter, is RhoC GTPase uitgedrukt in uitgezaaide kankercellen op relatief lage niveaus. Daarom hebben weinig RhoC-specifieke reagentia ontwikkeld. We hebben een aangepast Rho activatie test om RhoC GTPase op te sporen. Deze techniek maakt gebruik van een GST-Rho bindende domein fusie-eiwit te trekken actief RhoC GTPase. Daarnaast kunnen we totaal eiwit oogsten aan het begin van de test tot het niveau van het totaal (GTP en het BBP gebonden) RhoC GTPase te bepalen. Dit maakt voor de bepaling van actief versus het totaal RhoC GTPase in de cel. Een aantal commerciële versies van deze procedure zijn echter ontwikkeld, zijn de commerciële kits geoptimaliseerd voor RhoA GTPase en meestal niet goed werken voor RhoC GTPase. Delen van de test zijn eveneens gewijzigd als de ontwikkeling van een RhoC-specifiek antilichaam.
