<p> Belangrijke techinical punten zijn als volgt: (1) onderhoud van de fysiologische status van het dier (ventilatie, pH van de perfusative oplossing, lichaamstemperatuur), (2) injectie volume van de gebrandschilderde RBC, en (3) de voorwaarden voor observatie (optimale lens selectie, fluorescentie-intensiteit). Mogelijke toekomstige toepassingen zijn als volgt: (a) combinatie met pharmacophysiological en / of moleculaire biologische interventies, (b) lange termijn observatie van de arterioveneuze / arteriolo-sclerose en neovascularisatie. </p>

<p class="jove_content"> ACHTERGROND: Regulatory factoren en gedetailleerde fysiologie van in vivo microcirculatie zijn gebleven niet volledig duidelijk na vele verschillende modaliteiten van beeldvorming had uitgevonden. Terwijl veel macroscopische parameters van de bloedstroom reflecteren stroomsnelheid, veranderingen in de snelheid van de bloedstroom en rode bloedcellen (RBC) flux heeft lineaire relatie niet te houden in de microscopische waarnemingen. Er zijn meldingen van discrepantie tussen RBC snelheid en RBC flux, RBC flux en plasma flow volume, en van de ruimtelijke en temporele heterogeniteit van debietregeling in de perifere weefsels in microscopische waarnemingen, een wetenschappelijke basis voor de eis van meer gedetailleerde studies in microcirculatie regelgeving met behulp van intravitale microscopie. </p><p class="jove_content"> METHODEN: We gewijzigd Jeff Lichtman de methode van in vivo microscopische observatie van de muis musculus sternocleidomastoideus spieren. Muizen zijn verdoofd, geventileerd, en geïnjecteerd met PKH26L-fluorescent gelabelde rode bloedcellen voor microscopisch observatie. </p><p class="jove_content"> RESULTAAT &amp; CONCLUSIES: fluorescent gelabelde RBC worden gedetecteerd en goed herkenbaar aan een wide-field microscoop. Spiercontractie opgewekt door elektrische stimulatie geïnduceerde toename van RBC flux. Kwantificering van andere parameters zoals RBC snelheid en de capillaire dichtheid waren haalbaar zijn. Muizen goed verdragen de operatie, injectie van gebrandschilderd RBC, microscopische observatie, en elektrische stimulatie. Geen spieren of bloedvaten zijn aangetast werd waargenomen, wat suggereert dat onze methode is relatief minder invasieve en geschikt voor lange-termijn observaties. </p>
