<p> Wir bedanken uns bei JW Lichtman für seine Beratung in in vivo Beobachtung der Muskeln. </p>

<p class="jove_step"> RBC Membranfärbung </p><ol><li> Maus roten Blutkörperchen wurden aus dem gleichen Mausstamm mit Heparinisierung korrigiert. </li><li> Maus RBC wurden mit PKH26 Farbstoff (; Rot fluoreszierende Zelle Linker Kit, Sigma, USA 551/567 nm) gefärbt. </li><li> RBC wurden in PBS gewaschen und mit 2 um Lösung des PKH26 Farbstoff für 5 Minuten bei Raumtemperatur. <p> Wichtige techinical Punkte sind wie folgt: (1) Wartung der physiologische Zustand des Tieres (Lüftung, pH-Wert der Lösung perfusative, Körpertemperatur), (2) Injektionsvolumen des gefärbten RBC, und (3) Bedingungen für die Beobachtung (optimale Linse Auswahl-, Fluoreszenz-Intensität). Mögliche zukünftige Anwendungen sind wie folgt: (a) Kombination mit pharmacophysiological und / oder molekularbiologische Eingriffe, (b) langfristige Beobachtung der arterio / arteriolo-Sklerose und Gefäßneubildung. </p>

<p class="jove_content"> HINTERGRUND: regulatorische Faktoren und detaillierte Physiologie der In-vivo-Mikrozirkulation geblieben nicht in vollem Umfang nach vielen verschiedenen Modalitäten der Bildgebung erfunden geklärt. Während viele makroskopischer Parameter des Blutflusses Strömungsgeschwindigkeit, Änderungen in der Geschwindigkeit des Blutflusses und der roten Blutkörperchen (RBC) spiegeln Fluss nicht halten lineare Beziehung in die mikroskopischen Beobachtungen. Es gibt Berichte über Diskrepanz zwischen RBC Geschwindigkeit und RBC flux, flux RBC und Plasma-Volumenstrom und der räumlichen und zeitlichen Heterogenität der Durchflussregelung in den peripheren Geweben in mikroskopischen Beobachtungen, eine wissenschaftliche Grundlage für die Anforderung von detaillierteren Studien in der Mikrozirkulation der Verordnung über Intravitalmikroskopie. </p><p class="jove_content"> Methoden: Wir modifizierten Jeff Lichtman Methode der in vivo mikroskopische Beobachtung der Maus Kopfnicker Muskeln. Mäuse sind betäubt, belüftet, und injiziert mit PKH26L-fluoreszenzmarkierten Erythrozyten zur mikroskopischen Beobachtung. </p><p class="jove_content"> ERGEBNIS &amp; FAZIT: Fluoreszenz-markierte Erythrozyten werden erkannt und auch durch eine breite-Mikroskop aus. Muskelkontraktion durch elektrische Stimulation induzierte Zunahme RBC Flusses hervorgerufen. Quantifizierung von anderen Parametern, einschließlich RBC Geschwindigkeit und Kapillardichte möglich waren. Mäuse gut vertragen die Operation, Injektion von gefärbten roten Blutkörperchen, mikroskopische Beobachtung und elektrischer Stimulation. Kein Muskel-oder Blutgefäß beschädigt wurde beobachtet, was darauf hindeutet, dass unsere Methode relativ weniger invasive und geeignet für Langzeit-Beobachtungen ist. </p>
