Ett centralt tema i utvecklingsbiologi är diversifiering av linjerna och att klarlägga bakomliggande molekylära mekanismer. Detta innebär en grundlig analys av öden enskilda celler under normala och experimentella förhållanden. För detta ändamål transfektion metoder som riktas mot en enda stamceller är en förutsättning. Vi beskriver här en tekniskt enkel metod för transfecting enstaka celler i kyckling vävnader i-ovo, vilket gör att tillförlitliga härkomst spårning och genmanipulation. Specifik vävnad domäner är riktade i somit eller neuralrörsdefekter, och DNA injiceras direkt i epitelet av intresse, vilket resulterar i sporadiska transfektion av enskilda celler. Använda reportrar kan klonal populationer därmed spåras i upp till tre dagar, och beteendet hos genmanipulerade klonal populationer kan jämföras med det av kontrollerna. Denna metod utnyttjar tillgängligheten till kycklingembryo tillsammans med nya verktyg för genetisk manipulation. Vi jämför och diskutera dess fördelar jämfört med utbredda elektroporation metoden, och möjliga tillämpningar och användning i ytterligare in vivo-modeller är också föreslås. Vi förespråkar användandet av denna metod som ett viktigt tillskott och komplement till befintliga härstamning spåra och genetiska verktyg störningar.
