fabrication of micro-tissues using modules of collagen gel containing cells

Dieses Protokoll beschreibt die Herstellung von einer Art Mikro-Gewebe Module genannt. Die Modul-Ansatz generiert einheitliche, skalierbare und vaskularisierten Geweben. Die Module können von Kollagen sowie andere gelierbare oder vernetzbare Materialien hergestellt werden. Sie sind ca. 2 mm in der Länge und 0,7 mm Durchmesser auf Herstellung, sondern schrumpfen in Größe mit eingebetteten Zellen oder, wenn die Module sind mit Endothelzellen beschichtet. Die Module sind einzeln klein genug, dass die eingebetteten Zellen innerhalb der Diffusions-Limit von Sauerstoff und anderen Nährstoffen sind, sondern Module können zusammen verpackt werden, um größere Gewebe, perfundierbaren sind zu bilden. Diese Gewebe sind modular aufgebaut, da verschiedene Zelltypen in eingebettet werden kann oder beschichtet auf den Modulen, bevor sie zusammen, um komplexe Gewebe zu bilden verpackt werden. Es gibt drei wesentliche Schritte zu machen die Module: (1) Neutralisieren der Kollagen-und Einbettung Zellen in ihr, (2) Geliermittel das Kollagen in die Röhre und Schneiden der Module und (3) Beschichtung der Module mit Endothelzellen.
