<p class="jove_content">Kwantitatieve real-time PCR (QPCR) heeft ontpopt als een nauwkeurig en waardevol instrument in de profilering van genexpressie niveaus. Een van de vele voordelen is een lagere detectiegrens in vergelijking met andere methoden van gen expressie profilering tijdens het gebruik van kleinere hoeveelheden input voor elke test. Geautomatiseerde qPCR setup is verbeterd op dit gebied door toe te staan ​​voor een grotere reproduceerbaarheid. De handige en snelle setup zorgt voor high-throughput experimenten, waardoor de profilering van vele verschillende genen tegelijkertijd in elk experiment. Deze methode in combinatie met interne controle plaat vermindert ook de experimentele variabelen bij andere technieken. We hebben recent ontwikkelde een qPCR test voor profilering van pre-microRNA&#39;s (pre-miRNAs) met behulp van een set van 186 primer paren. MicroRNA hebben zich ontwikkeld tot een nieuwe klasse van kleine, niet-coderende RNA&#39;s met de mogelijkheid om vele doelen mRNA regelen bij de post-transcriptionele niveau. Deze kleine RNA&#39;s worden eerst getranscribeerd door RNA polymerase II als een primaire miRNA (pri-miRNA) transcriptie, die vervolgens wordt gesplitst in de voorloper miRNA (pre-miRNA). Pre-miRNAs worden geëxporteerd naar het cytoplasma waar Dicer knipt de hairpin loop te geven volwassen miRNAs. Verhogingen van miRNA niveaus kunnen worden waargenomen op zowel de voorloper en volwassen miRNA niveaus en profilering van beide vormen kan nuttig zijn. Er zijn verschillende commercieel verkrijgbare assays voor de rijpere miRNAs, maar de hoge kosten kunnen afschrikken onderzoekers van deze profilering techniek. Hier bespreken we een rendabele, betrouwbare, SYBR-based qPCR methode van profiling pre-miRNAs. Veranderingen in de pre-miRNA het niveau vaak een weerspiegeling van volwassen miRNA veranderingen en kan een nuttige indicator van de volwassen miRNA expressie. Kan echter gelijktijdig profilering van zowel pre-miRNAs en volwassen miRNAs worden optimaal als ze kunnen nonredundant informatie bijdragen en geven inzicht in de verwerking van microRNA. Bovendien kan de techniek hier beschreven worden uitgebreid met de profilering van andere bibliotheek sets voor specifieke studierichtingen of ziekteverwekkers omvatten.</p>
