De assays hierboven beschreven zijn die in verschillende studies het aantonen van de rol van meerdere factoren betrokken bij de vorming grens tussen de Schwann-astrocyten en beperkte Schwann cel migratie in astrocytaire omgeving. Schwann-cellen zijn een van de meest gebruikte cellen in reparatie strategieën volgen ruggenmergletsels. Schwann-cellen zijn in staat de ondersteuning van axonale regeneratie en ontspruiten door het afscheiden van groeifactoren 1,2 en het verstrekken van de groei bevorderen van adhesiemoleculen 3 en extracellulaire matrix moleculen 4. Bovendien zijn ze myelinate de gedemyeliniseerde axonen op de site van letsel 5. Echter na transplantatie, moet Schwann-cellen niet migreren van de site van implantaat en niet vermengen met de gastheer astrocyten 6,7. Dit resulteert in de vorming van een scherpe grens tussen de Schwann-cellen en astrocyten, een obstakel vormt voor het kweken van axonen proberen om het transplantaat weer af te sluiten in de gastheerweefsel proximaal en distaal. Astrocyten in contact met de Schwann-cellen ook ondergaan hypertrofie en up-reguleren van de remmende moleculen 8-13. In vitro testen zijn gebruikt om het model Schwann cel-astrocyten interacties en zijn belangrijk in het begrijpen van het mechanisme dat ten grondslag de cellulaire gedrag. Deze in vitro testen omvatten grens assay, waarbij een co-cultuur is gemaakt met behulp van twee verschillende cellen met elk celtype bezettende verschillende gebieden met slechts een kleine kloof tussen de twee fronten cel. Omdat de cellen delen en migreren, de twee mobiele fronten komen dichter bij elkaar en uiteindelijk botsen. Dit maakt het gedrag van de twee mobiele populaties te analyseren aan de grens. Een andere variatie van dezelfde techniek is om de twee mobiele populaties in cultuur mix en na verloop van tijd de twee soorten cellen te scheiden met Schwann-cellen samengeklonterd samen als eilanden in tussen astrocyten samen het creëren van meerdere Schwann-astrocyten grenzen. De tweede test wordt gebruikt in het bestuderen van de interactie van twee celtypen is de migratie test waar de cellulaire bewegingen kunnen worden gevolgd op het oppervlak van het andere celtype monolaag 14,15. Deze test is algemeen bekend als omgekeerde dekglaasje test. Schwann-cellen worden gekweekt op kleine glasdeeltjes en ze zijn omgekeerd naar beneden op het oppervlak van astrocyte monolagen en migratie wordt beoordeeld vanuit de rand van het dekglaasje. Beide testen zijn geweest bij het bestuderen van de onderliggende mechanismen die betrokken zijn bij de cellulaire uitsluiting en grens vorming. Een deel van de moleculen geïdentificeerd met behulp van deze technieken zijn N-cadherines 15, chondroïtinesulfaat proteoglycanen (CSPGs) 16,17, FGF / Heparine 18, Ef / Ephrins 19.
