ex vivo expansion of tumor-reactive t cells by means of bryostatin 1/ionomycin and the common gamma chain cytokines formulation

Er werd gemeld dat borstkanker bij pre-bestaande immuunrespons tegen hun tumoren 1,2 te hebben. Echter, een dergelijke immuunreacties geen volledige bescherming bieden tegen de ontwikkeling of herhaling van borstkanker. Om dit probleem te verhelpen door het verhogen van de frequentie van de tumor-reactieve T-cellen, is adoptieve immunotherapie in dienst. Een verscheidenheid aan protocollen die zijn gebruikt voor de uitbreiding van tumor-specifieke T-cellen. Deze protocollen zijn echter beperkt tot het gebruik van tumor antigenen ex vivo voor de activatie van antigeen-specifieke T-cellen. Hier presenteren wij een protocol voor de selectieve activering en uitbreiding van tumor-reactieve T-cellen uit de FVBN202 transgene muis-model van HER-2/neu positief borstcarcinoom voor gebruik in de adoptieve T cel-therapie van borstkanker. Het protocol bevat activatie van T-cellen met bryostatin-1/ionomycin (B / I) en IL-2 in de afwezigheid van tumor antigenen gedurende 16 uur. B / I activering nabootst intracellulaire signalen die resulteren in een T-cel activatie door het verhogen van proteïne kinase C-activiteit en intracellulair calcium, respectievelijk 4. Dit protocol activeert specifiek tumor-specifieke T-cellen, terwijl het doden van niet ter zake T-cellen. De B ​​/ I-geactiveerde T-cellen worden gekweekt met IL-7 en IL-15 voor 24 uur en dan gepulst met IL-2. Na 24 uur, T-cellen worden gewassen, splitsen en gekweekt met IL-7 + IL-15 voor extra 4 dagen.
