Les parasites protozoaires sont parmi les agents infectieuses les plus dévastatrices de l&#39;homme responsable d&#39;une variété de maladies, notamment l&#39;amibiase, qui est l&#39;une des trois causes les plus fréquentes de décès par maladie parasitaire. L&#39;agent de l&#39;amibiase est l&#39;amibe parasite Entamoeba histolytica qui existe sous deux étapes: le kyste infectieux présents dans les aliments ou l&#39;eau et les vivants trophozoïte envahissantes dans l&#39;intestin. Les manifestations cliniques de l&#39;amibiase large d&#39;être asymptomatique à la colite abcès, la dysenterie ou le foie. E. histolytica est l&#39;un des rares du parasite unicellulaire, avec le 5-méthylcytosine (5mC) dans son génome. 1, 2 Il contient un ADN simple méthyltransférase, Ehmeth, qui appartient à la famille Dnmt2. 2 Un rôle pour Dnmt2 dans le contrôle des éléments répétitifs a été établi dans E. histolytica, 3 Dictyostelium discoideum 4,5 et la drosophile. 6 Nos travaux récents ont montré que Ehmeth méthylates ARNt Asp, et cette constatation indique que cette enzyme a un ADN double / ARNt Asp activité méthyltransférase. 7 Cette observation est en accord avec la double activité qui a été rapporté pour D. discoideum et D. melanogaster. 8 La signification fonctionnelle de la spécificité ADN / ARNt de Dnmt2 enzymes est encore inconnue. Pour répondre à cette question, une méthode pour déterminer l&#39;ARNt activité méthyltransférase d&#39;Dnmt2 protéines a été établie. Dans cette vidéo, nous décrivons une approche simple pour préparer un substrat adéquat pour Dnmt2 ARNt et une méthode pour mesurer son activité ARNt méthyltransférase.
