We hebben een microfluïdische apparaat dat de levering en de systemische klaring van geneesmiddelen voor heterogene driedimensionale tumorweefsels in vitro nabootst. Voedingsstoffen die door vasculatuur niet alle delen van tumoren te bereiken, die tot heterogene micromilieus bestaande uit levensvatbare, rustige en necrotische celtypen. Veel kanker drugs niet effectief doordringen en behandelen alle soorten cellen hierdoor heterogeniteit. Monolagen van kankercellen niet bootsen deze heterogeniteit, waardoor het moeilijk om kanker farmaca met een geschikt in vitro model. Onze microfluïdische apparaten werden vervaardigd uit PDMS met behulp van zachte lithografie. Multicellulaire tumor sferoïden gevormd door de opknoping neerzetten, werden ingevoegd en beperkt tot rechthoekige kamers van het apparaat en onderhouden met continue perfusie medium aan een zijde. De rechthoekige vorm van de kamers van het apparaat lineaire gradiënten ontstaan ​​in weefsel. Fluorescerende vlekken werden gebruikt om e kwantificerene variabiliteit in apoptosis in weefsel. Tumoren van het apparaat werden behandeld met de fluorescerende chemotherapeuticum doxorubicine, time-lapse microscopie werd gebruikt om de verspreiding te controleren in weefsel en de effectieve diffusiecoëfficiënt geschat.
