Das retinale Pigmentepithel (RPE) liegt an der Rückseite des Säugetier-Auge, gerade unter der neuralen Netzhaut, die Photorezeptoren (Stäbchen und Zapfen) enthält. Das RPE ist eine Monoschicht von pigmentierten kubischen Zellen und Mitarbeiter eng mit der neuronalen Retina knapp darüber. Dieser Verein macht die RPE von großem Interesse für Forscher untersuchen Netzhauterkrankungen. Das RPE ist auch der Ort einer in vivo-Assay von Homologie-directed DNA-Reparatur, die p un-Assay. Die Maus Auge ist besonders schwer zu sezieren aufgrund seiner geringen Größe (ca. 3,5 mm im Durchmesser) und seine Kugelform. Dieser Artikel zeigt im Detail ein Verfahren für die Präparation des Auges führt einen ganzen Berg der RPE. In diesem Verfahren zeigen wir, wie man mit und nicht gegen die kugelförmige Struktur des Auges zu arbeiten. Kurz gesagt, sind das Bindegewebe, Muskeln und Sehnerv aus dem hinteren Teil des Auges entfernt. Dann werden die Hornhaut und die Linse entfernt. Als nächstes sind strategische Schnitte, die zu erheblichen Abflachung der übrigen Gewebe. Schließlich ist der neuronalen Retina vorsichtig abgehoben, enthüllt eine intakte RPE, die noch auf die zugrunde liegende Ader-und Lederhaut befestigt wird. Das ganze Berg kann die p un-Assay oder für die Immunhistochemie oder Immunfluoreszenz Beurteilung des RPE Gewebe durchzuführen.
