Den pigmentepitelet (RPE) ligger på baksidan av däggdjur ögat, precis under neurala näthinnan, som innehåller fotoreceptorer (stavar och tappar). RPE är ett monolager av pigmenterade cuboidal celler och medarbetare i nära samarbete med neurala näthinnan precis ovanför det. Denna förening gör RPE av stort intresse för forskare som studerar retinala sjukdomar. RPE är också platsen för en in vivo-test av homologi riktad DNA-reparation, p un-analysen. Musen ögat är särskilt svår att dissekera på grund av sin ringa storlek (ca 3,5 mm i diameter) och dess sfäriska form. Denna artikel visar i detalj ett förfarande för dissektion av ögat vilket resulterar i ett helt berg RPE. Vid detta förfarande visar vi hur du arbetar med, snarare än mot, den sfäriska struktur i ögat. Kortfattat, är bindväv, muskler och synnerven bort från baksidan av ögat. Då är hornhinnan och linsen tas bort. Därefter är strategiska nedskärningar som leder till betydande utplaning av de återstående vävnad. Slutligen är neurala näthinnan försiktigt lyftas av, avslöjar en intakt RPE, som fortfarande är knuten till den underliggande åderhinnan och sklera. Hela fäste kan användas för att utföra p un-analysen eller för immunohistokemi eller immunofluorescens bedömning av RPE vävnad.
