In dit artikel presenteren we een gedetailleerd protocol van monstervoorbereiding en driedimensionale reconstructie van de exosome complex met behulp van negatieve kleuring elektronenmicroscopie. Enkel deeltje elektronenmicroscopie (EM) reconstructie heeft onlangs uitgegroeid tot een populaire tool om de drie-dimensionale (3D) structuur van grote macromoleculaire complexen te krijgen. Vergeleken met X-ray kristallografie, heeft het een aantal unieke voordelen. Ten eerste, enkel deeltje EM reconstructie niet nodig om het eiwit monster, dat is het knelpunt in de X-ray kristallografie kristalliseren, vooral bij grote macromoleculaire complexen. Ten tweede is het niet nodig grote hoeveelheden eiwit monsters. Vergeleken met milligram van eiwitten die noodzakelijk zijn voor kristallisatie, enkel deeltje EM wederopbouw slechts enkele micro-liters van de eiwit-oplossing bij nano-molaire concentraties, het gebruik van de negatieve kleuring EM-methode. Ondanks een paar macromoleculaire assemblages met een hoge symmetrie, enkel deeltje EM is beperkt tot relatief lage resolutie (lager dan 1 nm resolutie) voor vele exemplaren in het bijzonder diegenen zonder symmetrie. Deze techniek wordt ook beperkt door de grootte van de moleculen onder studie, dat wil zeggen 100 kDa voor negatief gekleurde exemplaren en 300 kDa voor bevroren gehydrateerd exemplaren in het algemeen. Voor een nieuwe steekproef van onbekende structuur, we meestal gebruik van een heavy metal oplossing om de moleculen insluiten door negatieve kleuring. Het monster wordt vervolgens onderzocht in een transmissie-elektronenmicroscoop om tweedimensionale (2D) microfoto van de moleculen te nemen. In het ideale geval de eiwitmoleculen hebben een homogene 3D-structuur, maar vertonen verschillende oriëntaties in de microfoto. Deze microfoto zijn gedigitaliseerd en verwerkt in computers als &quot;single deeltjes&quot;. Met behulp van twee-dimensionale alignment en classificatie technieken, homogeen zijn moleculen in dezelfde standpunten geclusterd in klassen. Hun gemiddelde op verbetering van het signaal van 2D het molecuul vormen. Na wijzen we de deeltjes met de juiste relatieve oriëntatie (Euler hoeken), zullen we in staat zijn om de 2D-deeltje beelden te reconstrueren in een virtuele 3D-volume. In single deeltje 3D-reconstructie, een essentiële stap is het correct toewijzen van de juiste oriëntatie van elk afzonderlijk deeltje. Er zijn verschillende methoden om de weergave van elk deeltje toe te wijzen, waaronder de hoekige reconstitutie 1 en willekeurige conische tilt (RCT) methode 2. In dit protocol beschrijven we onze praktijk in het krijgen van de 3D-reconstructie van gist exosome complex met behulp van negatieve kleuring EM en RCT. Opgemerkt moet worden dat ons protocol van elektronen microscopie en beeldverwerking het basisprincipe van RCT volgt, maar is niet de enige manier om de methode uit te voeren. We beschrijven eerst hoe het eiwit monster in te bedden in een laag van Uranyl-Formate met een dikte vergelijkbaar met de grootte van eiwit, met behulp van een essen carbon rooster bedekt met een laag van continue dunne koolstof film. Dan het monster wordt in een transmissie-elektronenmicroscoop te verzamelen Untilted (0 graden) en gekanteld (55 graden) paren van microfoto die later zullen worden gebruikt voor de verwerking en het verkrijgen van een eerste 3D-model van de gist exosome. Daartoe voeren we RCT en verfijnen van de eerste 3D-model met behulp van de projectie-matching verfijning methode 3.
