Raman spectroscopie wordt vaak geplaagd door een sterke fluorescerende achtergrond, in het bijzonder voor biologische monsters. Als een monster wordt opgewekt met een trein van ultrasnelle pulsen, een systeem dat tijdelijk kan spectraal aparte overlappende signalen op een picoseconde tijdschaal kan isoleren tijdig aankomen Raman verstrooide licht van de late aankomst fluorescentie licht. Hier bespreken we de bouw en exploitatie van een complex niet-lineaire optische systeem dat alle-optisch schakelen gebruikt in de vorm van een low-power optische Kerr gate te isoleren Raman en fluorescentie-signalen. Een enkele 808 nm laser met 2,4 W van de gemiddelde vermogen en 80 MHz herhalingsfrequentie is gesplitst, met ongeveer 200 mW van 808 nm licht wordt omgezet tot &lt;5 mW van 404 nm licht naar het monster te Raman verstrooiing te wekken. De resterende onbekeerde 808 nm licht wordt vervolgens verstuurd naar een niet-lineaire medium waar het fungeert als de pomp voor de all-optische sluiter. De sluiter opent en sluit in 800 fs met een maximale efficiëntie van ongeveer 5%. Met behulp van dit systeem zijn wij in staat om succesvol Raman en fluorescentie-signalen gescheiden op een 80 MHz herhalingsfrequentie met behulp van pulse energieën en krachten die de gemiddelde blijven biologisch veilig. Omdat het systeem geen extra capaciteit in termen van optische macht heeft, hebben we een aantal detail ontwerpen en de afstemming overwegingen die helpen bij het maximaliseren van de productiviteit van het systeem. We bespreken ook ons ​​protocol voor het verkrijgen van de ruimtelijke en temporele overlap van het signaal en de pomp balken in het Kerr medium, evenals een gedetailleerd protocol voor de spectrale acquisitie. Tot slot melden wij een enkele representatieve resultaten van Raman-spectra verkregen in de aanwezigheid van sterke fluorescentie met behulp van onze tijd-gating systeem.
