Gevestigde cellijnen zijn een kritisch onderzoek tool die het gebruik van proefdieren kunnen verminderen in het onderzoek. Bepaalde stammen van genetisch gemodificeerde muizen, zoals ATM - / - en p53 - / - consequent te ontwikkelen thymus lymfoom vroeg in het leven 1,2, en dus kunnen dienen als een betrouwbare bron voor afleiding van muriene T-cellijnen. Hier presenteren we een gedetailleerd protocol voor de ontwikkeling van bestaande muizen thymus lymfoom T-cellijnen zonder de noodzaak om interleukines zoals beschreven in de vorige protocollen 1,3 toe te voegen. Tumoren werden geoogst uit muizen leeftijd van drie tot zes maanden, ten vroegste aanwijzing van zichtbare tumoren gebaseerd op de observatie van de gebogen houding, moeilijke ademhaling, slechte verzorging en verspillen in een vatbare stam 1,4. We hebben met succes een aantal T-cellijnen met behulp van dit protocol en inteeltstammen van ATM - / - [FVB/N- Atm tm1Led / J] 2 en p53 - / - [129/S6- Trp53 tm1Tyj / J] 5 muizen. We verder zien dat meer dan 90% van de gevestigde T-cel populatie CD3, CD4 en CD8 uitdrukt. In overeenstemming met stabiel gevestigde cellijnen, hebben de T-cellen gegenereerd met behulp van het huidige protocol is gepasseerd voor meer dan een jaar.
