De procedure hier gepresenteerde maakt gebruik van een Bis-Tris gel met MES draait buffer als een voorbeeld van de denaturering PAGE het gebruik van de Invitrogen NuPAGE Novex elektroforese-systeem. Western Blotting (of immunoblotting) is een standaard laboratorium procedure die de onderzoekers om de expressie van een eiwit te controleren, bepalen de relatieve hoeveelheid van het eiwit aanwezig is in verschillende monsters en analyseren van de resultaten van co-immunoprecipitatie experimenten. In deze methode, is een doelwit eiwit gedetecteerd met een specifiek primair antilichaam in een bepaald monster van weefsel homogenaat of het uittreksel. Eiwit scheiding volgens moleculair gewicht is bereikt door het gebruik denaturerende SDS-PAGE. Na overdracht aan een membraan, is het doelwit eiwit gemerkt met een specifiek primair antilichaam en gedetecteerd door chemiluminescentie. Sinds de eerste beschrijving, heeft de westerse-blotting techniek ondergaan verschillende verbeteringen, waaronder pre-cast gels en gebruiksvriendelijke apparatuur. In ons laboratorium hebben we gekozen voor de commercieel beschikbare NuPAGE elektroforese systeem van Invitrogen. Het is een innovatief neutrale pH, discontinue SDS-PAGE, pre-cast mini-gel systeem. Dit systeem biedt verschillende voordelen ten opzichte van de traditionele Laemmli techniek zoals: i) een langere houdbaarheid van de pre-cast gels, variërend van 8 maanden tot 1 jaar; ii) een brede scheiding reeks molecuulgewichten 1 tot 400 kDa, afhankelijk van het type van de gel gebruikt, en iii) een grotere veelzijdigheid (range van acrylamide percentage, de aard van de gel, en de ionische samenstelling van de lopende buffer). De procedure zoals beschreven in deze video artikel maakt gebruik van de Bis-Tris discontinu buffersysteem met 4-12% Bis-Tris gradiënt gels en MES draaien buffer, als een illustratie van hoe een western blot-uit te voeren met behulp van de Invitrogen NuPAGE elektroforese-systeem. In ons laboratorium hebben wij goede en reproduceerbare resultaten voor diverse biochemische toepassingen het gebruik van deze western-blotting methode.
