La procédure présentée ici utilise un gel de Bis-Tris avec MES Tampon comme un exemple de dénaturation PAGE en utilisant le système d&#39;Invitrogen NuPAGE électrophorèse Novex. Western Blot (ou immuno-) est une procédure standard de laboratoire permettant aux enquêteurs de vérifier l&#39;expression d&#39;une protéine, de déterminer la quantité relative de la protéine présente dans les différents échantillons, et d&#39;analyser les résultats des expériences de co-immunoprécipitation. Dans cette méthode, une protéine cible est détectée avec un anticorps primaire spécifique dans un échantillon donné d&#39;homogénat de tissu ou de l&#39;extrait. Séparation des protéines selon leur poids moléculaire est réalisé en utilisant dénaturant SDS-PAGE. Après transfert sur une membrane, la protéine cible est sondée avec un anticorps spécifique primaire et détecté par chimioluminescence. Depuis sa première description, la technique de western-blot a subi plusieurs améliorations, dont préfabriqués gels et conviviale de l&#39;équipement. Dans notre laboratoire, nous avons choisi d&#39;utiliser le système NuPAGE commercialement disponible chez Invitrogen électrophorèse. Il s&#39;agit d&#39;un pH neutre, innovant, discontinue SDS-PAGE, préfabriqués mini-système de gel. Ce système présente plusieurs avantages par rapport à la technique traditionnelle de Laemmli notamment: i) une durée de conservation des gels préfabriqués allant de 8 mois à 1 an; ii) une gamme de séparation éventail de poids moléculaires de 1 à 400 kDa selon le type du gel utilisé, et iii) une plus grande polyvalence (gamme de pourcentage d&#39;acrylamide, le type de gel, et la composition ionique du tampon d&#39;exécution). La procédure décrite dans cet article la vidéo utilise le système de tampon Bis-Tris discontinue avec 4-12% de Bis-Tris gels de gradient et MES courir tampon, comme une illustration de la façon d&#39;effectuer un test western blot-en utilisant le système d&#39;Invitrogen électrophorèse NuPAGE. Dans notre laboratoire, nous avons obtenu de bons résultats et reproductible pour diverses applications biochimiques en utilisant cette méthode western-blot.
