O procedimento sugerido aqui usa um gel Bis-Tris com MES tampão de corrida como um exemplo de desnaturação PAGE utilizando o sistema de eletroforese Invitrogen NuPAGE Novex. Western Blot (ou immunoblotting) é um procedimento padrão de laboratório permitindo que pesquisadores para verificar a expressão de uma proteína, determinar a quantidade relativa das proteínas em diferentes amostras e analisar os resultados da co-imunoprecipitação experimentos. Neste método, uma proteína-alvo é detectado com um anticorpo específico primária em uma dada amostra de homogeneizado de tecido ou extrato. Separação de proteínas de acordo com o peso molecular é obtida através de desnaturação SDS-PAGE. Após a transferência para uma membrana, a proteína-alvo é sondado com um anticorpo primário específico e detectados por quimioluminescência. Desde sua primeira descrição, a técnica de western-blotting passou por várias melhorias, incluindo a pré-cast géis e user-friendly equipamento. Em nosso laboratório, optou-se por utilizar o sistema de eletroforese comercialmente disponíveis NuPAGE da Invitrogen. É um pH neutro inovadoras, descontínua SDS-PAGE, pré-moldados sistema de mini-gel. Este sistema apresenta várias vantagens sobre a técnica de Laemmli tradicionais, incluindo: i) uma vida útil mais longa dos géis pré-moldados que variam de 8 meses a 1 ano; ii) uma ampla gama de separação pesos moleculares 1-400 kDa, dependendo do tipo de gel utilizados, e iii) maior versatilidade (variação de porcentagem de acrilamida, o tipo de gel, e da composição iônica do tampão de corrida). O procedimento descrito neste artigo utiliza o vídeo Bis-Tris sistema tampão descontínuo com 4-12% Bis-Tris gradiente géis e MES tampão de corrida, como uma ilustração de como realizar um western blot usando o sistema de eletroforese Invitrogen NuPAGE. Em nosso laboratório, temos obtido bons resultados e reprodutível para várias aplicações bioquímicas usando este método western-blotting.
