Förfarandet som presenteras här används en Bis-Tris gel med MES löpande buffert som ett exempel på denaturering sidan med hjälp av Invitrogen NuPAGE Novex elektrofores system. Dessutom ger detta prefabricerade gel systemet protein separation i denaturering eller icke-denaturering villkor samt rymmer ett brett spektrum av molekylvikt (1 till 200 kDa för BIS-Tris geler till 36-400 kDa för Tris- Acetat geler). Western blotting (eller immunoblotting) är en standard laboratorium förfarande som medger utredare för att kontrollera uttrycket av ett protein, bestämma den relativa mängden av det protein som finns i olika prover och analysera resultaten av samarbetet immunoprecipitation experiment. I denna metod är ett målprotein upptäckas med en specifik primära antikroppen i ett visst prov av vävnad homogenatet eller utdraget. Protein separation enligt molekylvikt uppnås med hjälp av denaturering SDS-PAGE. Efter överföring till ett membran, är det målprotein sonderade med en specifik primär antikropp och upptäcks av kemiluminiscens. Sedan den första beskrivningen, har den västra-analys tekniken genomgått flera förbättringar, inklusive prefabricerade geler och användarvänlig utrustning. I vårt laboratorium har vi valt att använda kommersiellt tillgängliga systemet NuPAGE elektrofores från Invitrogen. Det är ett innovativt neutralt pH, diskontinuerlig SDS-PAGE, pre-cast mini-gel-system. Detta system ger flera fördelar jämfört med de traditionella Laemmli tekniken inbegripet i) en längre hållbarhet av prefabricerade geler från 8 månader till 1 år, ii) en bred separation rad molekylvikter 1 till 400 kDa beroende på typ av gel som används, och iii) större flexibilitet (utbud av akrylamid procent, typ av gel, och det joniska sammansättningen av löpande buffert). Det förfarande som beskrivs i den här videon artikeln utnyttjar Bis-Tris diskontinuerlig buffert system med 4-12% Bis-Tris gradient geler och MES löpande buffert, som en illustration till hur man utför en västerländsk blot med Invitrogen systemet NuPAGE elektrofores. I vårt laboratorium har vi fått goda och reproducerbara resultat för olika biokemiska tillämpningar med denna western-analys metod.
