studying cell rolling trajectories on asymmetric receptor patterns

Zijdelingse verplaatsing van cellen loodrecht op een stroom stroom door walsen op asymmetrische receptor patronen biedt een gelegenheid voor de ontwikkeling van nieuwe apparaten voor de label-free scheiding en analyse van cellen 1. Dergelijke apparaten kunnen gebruik maken van zijdelingse verplaatsing voor continue-flow scheiding, of receptor patronen die hechting moduleren om onderscheid te maken tussen de verschillende cel fenotypes of niveaus van receptor expressie. Begrijpen van de aard van de cel rollen trajecten op receptor-patroon substraten is noodzakelijk voor de engineering van de substraten en het ontwerp van dergelijke apparaten. Hier tonen we een protocol voor het bestuderen van cellen rollen trajecten op asymmetrische receptor patronen die cel rollen adhesie 2 ondersteunen. Goed gedefinieerde, um-schaal patronen van P-selectine-receptoren werden vervaardigd met behulp van microcontact afdrukken op goud gecoate dia&#39;s die werden opgenomen in een flow kamer. HL60 cellen die de PSGL-1-ligand 3 werden stroomde over een gebied van de patroon lijnen en gevisualiseerd op een omgekeerde helder veld microscoop. De cellen gerold en gevolgd langs de schuine randen van de patronen, wat resulteert in een zijwaartse afbuiging. Elke cel meestal van staal voor een bepaalde afstand langs de randen patroon (gedefinieerd als de rand tracking lengte), los van de rand, en opnieuw aan een downstream patroon. Hoewel dit detachement maakt het moeilijk om volgen het gehele traject van een cel van de ingang af te sluiten in de stroom kamer, was particle-tracking-software gebruikt om te analyseren en de glooiende trajecten van de cellen opbrengst in de tijd toen ze bewegen op een enkele receptor -patroon lijn. De trajecten werden vervolgens onderzocht op uitkeringen van cel-rollen snelheden en de rand tracking lengtes voor elke cel voor verschillende patronen te verkrijgen. Dit protocol is handig voor het kwantificeren van cel-rollen trajecten op de receptor patronen en met betrekking tot deze technische parameters zoals hoek van het patroon en de shear stress. Deze gegevens zal nuttig zijn voor het ontwerp van microfluïdische apparaten voor het label-free cell scheiding en analyse.
