Der Einfluss der Stroma-Zellen, einschließlich Fibroblasten auf Mamma-Tumorprogression wurde auch durch den Einsatz von Maus-Modellen dokumentiert, insbesondere durch die Transplantation von Stromazellen und Epithelzellen in der Brustdrüse von Mäusen. Das Fehlen von funktionellen T-Zellen aus Mäusen Immunsystem verhindert eine genaue Berechnung der Stromazellen auf Mamma Tumorprogression in vivo, mit wichtigen Auswirkungen auf die Entwicklung von Medikamenten und Wirksamkeit. Darüber hinaus sind immungeschwächte Mäuse teuer, schwer zu züchten und bedürfen einer besonderen Pflege Bedingungen. Um diese Hindernisse zu überwinden, haben wir einen Ansatz zur orthotop transplantierten Stromazellen und Epithelzellen in Mäuse aus dem gleichen genetischen Hintergrund, um konsistente Tumorbildung induzieren entwickelt. Dieses System beinhaltet die Ernte normal, Karzinom assoziiert Fibroblasten, PyVmT Mammakarzinom-Zellen und Kollagen vom Spender C57BL/6J-Mäuse. Die Zellen werden dann in Kollagen eingebettet und transplantiert in der Leistengegend Milchdrüsen der weiblichen C57BL/6J-Mäuse. Die Transplantation von Zellen allein PyVmT Form tastbaren Tumoren 30-40 Tage nach der Transplantation. Während Zellen und Matrix aus C57BL/6J-Mäuse in diesen Studien verwendet wurden, kann die Isolierung von Zellen und Matrix und Transplantation Ansatz Mäuse aus verschiedenen genetischen Hintergründen demonstriert Vielseitigkeit angewendet werden.
