1. Representatieve resultaten: Een schema van onze FDA-goedgekeurde multivirus-specifieke CTL generatie proces is weergegeven in figuur 1. In tegenstelling tot de conventie multivirus CTL protocollen die adenovectors en EBV-LCL te gebruiken om virus-reactieve T-cellen te stimuleren 2 hebben we infectieus virus materiaal vervangen door DNA plasmiden die coderen voor meerdere antigenen afgeleid van elk van de virussen 3. Het stimuleren van trivirus CTL ontwierpen we drie multicistronic plasmiden die coderen voor Hexon en Penton van adenovirus, IE1 en pp65 van CMV, en EBNA1, LMP2, en BZLF1 van EBV. Verder te optimaliseren onze productie-methoden die we samen met de Natuur-technologie, die geminimaliseerd, antibiotica-vrij (FDA-compatibel) plasmiden gegenereerd voor CTL stimulatie 13,14. Met behulp van deze strategie die we consequent te bereiken nucleofection efficiëntie van&gt; 35% met behoud van hoge levensvatbaarheid van de cellen (gegevens niet getoond) 3. Figuur 2 blijkt dat de frequentie van de virus-specifieke T-cellen in reactie op geoptimaliseerde DNA plasmiden zoals gemeten door IFNy ELISPOT, was groter dan in de respons op de conventionele pShuttle-based expressieplasmiden de uiting van de dezelfde antigenen (n = 8 Adenovirus, n = 4 CMV, en n = 2 EBV). Virale infecties veroorzaken morbiditeit en mortaliteit bij allogene hematopoietische stamceltransplantatie (HSCT) ontvangers. Wij en anderen hebben met succes gegenereerd en toegediende T-cellen die specifiek zijn voor het Epstein Barr virus (EBV), cytomegalovirus (CMV) en adenovirus (Adv) met behulp van monocyten en EBV getransformeerde lymfoblastoïde cel (EBV-LCL) gen-gemodificeerd met een adenovirus vector als antigeen presenterende cellen (APC&#39;s). Zo weinig als 2x10 5 / kg trivirus-specifieke cytotoxische T lymfocyten (CTL) zich verspreid door verschillende logs na infusie en leek te voorkomen en te behandelen, zelfs een ernstige virale ziekte resistent zijn tegen andere beschikbare therapieën.
