Hemagglutinine (HA) en neuraminidase (NA) zijn twee oppervlakte-eiwitten van influenza virussen waarvan bekend is dat een belangrijke rol spelen in de virale levenscyclus en de inductie van beschermende immuunrespons 1,2. Aangezien de belangrijkste doelstelling voor neutraliserende antistoffen, wordt HA momenteel gebruikt als het griepvaccin potentie marker en wordt gemeten door een radiale immunodiffusie (SRID) 3. Echter, de afhankelijkheid van SRID op de beschikbaarheid van de overeenkomstige subtype-specifieke antisera zorgt voor een minimum van 2-3 maanden vertraging voor de introductie van elke nieuwe vaccin. Bovendien, ondanks de bewijzen dat NA ook beschermende immuniteit 4, het aantal NA in griepvaccins is nog niet gestandaardiseerd te wijten aan een gebrek aan geschikte reagentia of analysemethode 5 induceert. Zo simpel alternatieve methoden in staat te kwantificeren HA en NA antigenen zijn wenselijk voor een snelle release en een betere kwaliteitscontrole van griepvaccins. Universeel geconserveerde gebieden in alle beschikbare influenza-A-HA en NA sequenties werden geïdentificeerd door bioinformatica analyses 6-7. Een sequence (aangeduid als Uni-1) werd geïdentificeerd in het enige universeel bewaard epitoop van HA, de fusiepeptide 6, terwijl de twee geconserveerde sequenties werden geïdentificeerd in neuraminidases, een dicht bij de enzymatische actieve site (aangeduid als HCA-2) en de andere dicht bij de N-terminus (aangeduid als HCA-3) 7. Peptiden met deze aminozuursequenties werden gesynthetiseerd en gebruikt om konijnen voor de productie van antilichamen immuniseren. Het antilichaam tegen de Uni-1 epitoop van HA in staat was om te binden tot en met 13 subtypen van het influenza A HA (H1-H13), terwijl de antistoffen tegen de HCA-2 en HCA-3 regio&#39;s van NA in staat waren bindend voor alle negen NA subtypes. Alle antilichamen toonde opmerkelijke specificiteit tegen de virale sequenties, zoals blijkt uit de constatering dat er geen kruisreactiviteit met eiwitten allantoïsche was gedetecteerd. Deze universele antilichamen werden vervolgens gebruikt om slot blot assays te ontwikkelen voor HA en NA in het influenza-A-vaccins te kwantificeren, zonder de noodzaak van specifieke antisera 7,8. Vaccin monsters werden aangebracht op een PVDF-membraan met behulp van een slot blot apparaat samen met de referentie-normen verdund tot verschillende concentraties. Voor de detectie van HA, werden monsters en de standaard eerste verdund in Tris-gebufferde zoutoplossing (TBS) met 4M ureum, terwijl voor het meten van de NA ze werden verdund in TBS met 0,01% Zwittergent als deze voorwaarden significant de detectie gevoeligheid verbeterd. Na de vaststelling van de HA en NA antigenen door immunoblotting met hun respectieve universele antilichamen werden signaal intensiteiten gekwantificeerd door densitometrie. Bedragen van de HA en NA in de vaccins werden vervolgens berekend met behulp van een standaardcurve opgesteld met het signaal intensiteit van de verschillende concentraties van de gebruikte referenties. Gezien het feit dat deze antilichamen binden aan universele epitopen in HA of NA, kunnen geïnteresseerde onderzoekers gebruiken als research tools in immunoassays andere dan de sleuf blot alleen.
