Bacteriële oppervlakte-eiwitten betrokken zijn bij direct contact met gastheercellen en in de opname van voedingsstoffen uit het milieu 1. Om deze reden kan cellulaire lokalisatie geven inzicht in de functionele rol van bacteriële eiwitten. Oppervlak lokalisatie van bacteriële eiwitten is een belangrijke stap op weg naar de identificatie van virulentiefactoren betrokken bij mechanismen van pathogeniteit. Methoden voor het fractioneren leptospiral membranen 02-05 mei selectief zijn voor een bepaalde klasse van de buitenste membraan-eiwitten (Omps), zoals lipoproteïnen versus transmembrane Omps, en dus leiden tot misclassificatie. Dit waarschijnlijk is te wijten aan structurele verschillen en hoe ze worden geassocieerd met de buitenste membraan. Lipoproteïnen worden geassocieerd met membranen via een hydrofobe interactie tussen de N-terminale lipide-groep (drie vetzuren) en het lipide bilaag fosfolipiden 6, 7. In tegenstelling, zijn transmembraan Omps meestal geïntegreerd in de lipide bilaag door amfipathisch β-sheets die in een vat-achtige structuur 8, 9. Bovendien betekent de aanwezigheid van een eiwit in de buitenste membraan niet per se garanderen dat het eiwit of haar domeinen worden blootgesteld aan de oppervlakte. Spirochetal buitenste membranen is bekend dat ze kwetsbaar en dit betekent dat methoden waarbij zachte manipulatie van cellen en opneming van sub-oppervlakte-eiwit regelt het beoordelen van de integriteit van de buitenste membraan. Hier presenteren we een immunofluorescentie assay (IFA) methode om rechtstreeks te beoordelen oppervlak blootstelling van eiwitten op intact leptospiren. Deze methode is gebaseerd op de erkenning van leptospiral oppervlakte-eiwitten door antigeen-specifieke antilichamen. Hierin worden antilichamen specifiek voor OmpL54 10 detetcted aftero binding aan native, het oppervlak blootgesteld epitopen. Vergelijking van antilichaamreactiviteit intact versus gepermeabiliseerde cellen in staat stelt de evaluatie van de cellulaire distributie en het al dan niet een eiwit selectief aanwezig is op leptospiral oppervlak. De integriteit van de buitenste membraan moet worden beoordeeld met behulp van antilichamen tegen een of meer ondergrondse proteïnen, bij voorkeur gelegen in het periplasma. Het oppervlak IFA-methode kan worden gebruikt om het oppervlak blootstelling van elke leptospiral eiwit waaraan specifieke antilichamen beschikbaar te analyseren. Zowel het nut en de beperking van de methode hangt af van de vraag of de gebruikte antilichamen kunnen binden de inheemse epitopen. Omdat antilichamen vaak worden opgewekt tegen recombinante eiwitten, epitopen van inheemse, oppervlakte-eiwitten kunnen worden blootgesteld niet worden herkend. Toch is het oppervlak IFA-methode is een waardevol instrument voor het bestuderen van onderdelen van intacte bacteriële oppervlakken. Deze methode kan worden toegepast, niet alleen voor leptospiren, maar ook andere spirocheten en gramnegatieve bacteriën. Voor een sterkere conclusies ten aanzien van oppervlakte-blootstelling van Omps, is een integrale aanpak, waarbij verschillende mobiele lokalisatie methoden aanbevolen 10.
