Eritropoiese é um sistema modelo comumente usado para estudar a diferenciação celular. Durante a eritropoiese, pluripotentes humano adulto células-tronco hematopoéticas (HSCs) diferenciar em progenitores oligopotent, precursores e glóbulos vermelhos maduros 1. Este processo é regulado por uma grande parte ao nível da expressão gênica, em que fatores de transcrição específicos ativar genes de linhagem específica, enquanto concomitantemente reprimindo genes que são específicos para outros tipos de células 2. Estudos sobre fatores de transcrição que regulam a eritropoiese são frequentemente realizadas usando linhas de células humanas e camundongos, que representam, até certo ponto, as células eritróides, em determinadas fases de diferenciação 3-5. Linhagens de células transformadas no entanto só pode imitar parcialmente células eritróides e mais importante é que eles não permitem que se compreensível estudar as mudanças dinâmicas que ocorrem como células progridem por muitos estágios para o seu destino eritróide final. Portanto, um desafio atual continua a ser o desenvolvimento de um protocolo para obter populações relativamente homogêneas de HSCs primária e células eritróides em vários estágios de diferenciação em quantidades que são suficientes para executar genômica e proteômica experimentos. Aqui nós descrevemos um ex vivo protocolo de cultura de células para induzir a diferenciação eritróide de humano-tronco hematopoiéticas / progenitoras que foram isolados a partir de qualquer sangue do cordão umbilical, na medula óssea ou sangue periférico de adultos mobilizados com G-CSF (leucaférese). Este sistema de cultivo, inicialmente desenvolvido pelo laboratório Douay 6, usa citocinas e co-cultura em células mesenquimais para imitar o microambiente da medula óssea. Usando este protocolo diferenciação ex vivo, observamos uma ampliação forte de progenitores eritróides, uma indução de diferenciação exclusivamente para a linhagem eritróide e uma maturação completa para o estágio de enucleados células vermelhas do sangue. Assim, este sistema oferece uma oportunidade para estudar o mecanismo molecular de regulação da transcrição como células-tronco hematopoiéticas progresso ao longo da linhagem eritróide. Estudando eritropoiese no nível de transcrição também requer a capacidade de sobre-expressar ou knockdown fatores específicos em células primárias eritróide. Para este fim, usamos um lentivírus mediada sistema de entrega de gene que permite a infecção eficiente de ambos dividindo e não dividir células 7. Aqui nós mostramos que somos capazes de forma eficiente o knockdown TAL1 fator de transcrição em células primárias de eritróide humana. Além disso, a expressão GFP demonstra uma eficiência de infecção lentiviral cerca de 90%.
