Erytropoes är ett vanligt använt modellsystem för att studera celldifferentiering. Under erytropoes pluripotenta vuxen människa hematopoetiska stamceller (HSCs) differentiera till oligopotent progenitorceller, engagerade prekursorer och mogna röda blodkroppar 1. Denna process regleras till stor del på nivån av genuttryck, där specifika transkriptionsfaktorer aktiverar härstamning-specifika gener medan samtidigt förtränga gener som är specifika för andra celltyper 2. Studier av transkriptionsfaktorer som reglerar erytropoes utförs ofta med humana och murina cellinjer som representerar i viss mån, erytroida celler vid givna stadier av differentiering 3-5. Men förvandlas cellinjer kan bara delvis härma erytroida celler och viktigast de inte tillåter en att begripligt studera de dynamiska förändringar som sker som celler framsteg genom många stadier mot deras slutgiltiga erytroida öde. Därför är fortfarande en aktuell utmaning att utveckla ett protokoll för att få relativt homogen befolkning av primär HSCs och erytroida celler i olika stadier av differentiering i tillräckligt stora mängder för att utföra genomik och proteomik experiment. Här beskriver vi en ex vivo-protokoll cellodling att framkalla erytroida differentiering från mänskliga hematopoetisk stam / progenitorceller som har isolerats från antingen navelsträngsblod, benmärg eller vuxna perifert blod mobiliserade med G-CSF (leukaferes). Denna kultur-systemet, som ursprungligen utvecklats av Douay laboratoriet 6, använder cytokiner och co-kultur på mesenkymala celler för att efterlikna mikromiljö benmärgen. Med denna ex protokoll vivo differentiering, observerar vi en stark förstärkning av erytroida progenitorceller, en induktion av differentiering uteslutande mot erytroida härstamning och en fullständig mognad till det stadium av enucleated röda blodkroppar. Således ger detta system en möjlighet att studera de molekylära mekanismen för transkriptionell reglering som hematopoetiska stamceller framsteg på erytroida härstamning. Studera erytropoes på transkriptionell nivå kräver också förmåga att över-express eller ÖVERVÄLDIGANDE specifika faktorer i primär erytroida celler. För detta ändamål använder vi en lentivirus-medierad systemet gen leverans som möjliggör en effektiv infektion av både dela och icke-delande celler 7. Här visar vi att vi kan effektivt ÖVERVÄLDIGANDE de TAL1 transkriptionsfaktor i primära mänskliga erytroida celler. Dessutom visar GFP uttryck en effektivitet Lentiviral infektion nästan 90%. Således ger vår protokollet ett mycket användbart system för karakterisering av den rättsliga nätverk av transkriptionsfaktorer som styr erytropoes.
