Explantatie elektroporatie is een eenvoudige manier te kwantificeren cis-regulerende activiteit in de zich ontwikkelende muis netvlies. In vergelijking met cis-regulatorische analyse via de muis transgenese, elektroporatie is veel goedkoper, is er slechts pasgeboren muis pups, DNA, ontleden instrumenten, en elektroporatie / weefselkweek apparatuur. Het is ook veel minder tijd in beslag: een experiment vereist slechts een paar uur van de voorbereidingstijd, een cultuur periode van ongeveer acht dagen, en een paar uur aan het eind van het experiment voor de beeldvorming en data-analyse. Explantatie elektroporatie is ook superieur aan celcultuur gebaseerde cis-regulatorische analyse omdat de werkelijke retinaal weefsel wordt gebruikt. Het netvlies ontwikkelt zich heel normaal in explantatie cultuur-it maakt drie verschillende cellulaire lagen (buitenste nucleaire laag, binnenste nucleaire laag en ganglion cellaag), hoewel fotoreceptoren niet buitenste segmenten uit te werken. Een ander voordeel is dat Explantatie elektroporatie is zeer reproduceerbaar. Dezelfde constructen geëlektroporeerd in verschillende netvlies, zelfs op verschillende dagen, meestal resulteert in dezelfde relatieve expressie niveaus. Aangezien de geëlektroporeerd plasmiden wordt gedacht dat episomally worden gehandhaafd in de kern en zijn niet opgenomen in de chromosomen, zij niet lijken te zijn onderworpen aan dezelfde integratie plaats effecten die cis-regulatorische analyse uitgevoerd in transgene muizen mee kampen. Explantatie elektroporatie heeft wel een aantal beperkingen. Ten eerste kan alleen cellen die nog in de cel cyclus efficiënt worden getransduceerd door elektroporatie 15. Bij P0, staven en andere later geboren netvlies celtypen (bipolaire cellen, amacrine cellen, M ller glia) zijn de belangrijkste celpopulaties het doelwit van deze methode. Elektroporatie van de kegel fotoreceptoren door P0 elektroporatie is gemeld, maar het rendement 16 lijkt laag te zijn. Een tweede beperking is dat explantatie cultuur buiten de twee weken resulteert in een progressieve afwijking van het netvlies en wordt daarom niet aanbevolen. Als promotor kwantificering is nodig op late tijdstippen, maar een in-vivo electroporatie 9 kan worden uitgevoerd, gevolgd door het netvlies dissectie op de gewenste tijdpunt, flat-montage van de ontleed netvlies, en kwantificering zoals beschreven in hoofdstuk 9. Een derde beperking is dat deze test is slechts matig high-throughput. In tegenstelling tot de celcultuur-gebaseerde testen die kunnen testen honderden constructen in een enkel experiment, de techniek beschreven in dit protocol vereist een minimum van een hele muis netvlies per bouwen. Zo kan slechts een paar dozijn construeert redelijkerwijs kan worden geëlektroporeerd in een dag. Een extra waarschuwing met betrekking tot de kwantificering van de promotor activiteit met behulp van de huidige aanpak is dat er een potentieel voor &#39;bloeden-through&#39; van DsRed fluorescentie in het GFP-kanaal, met name bij het testen van een zeer sterke promotors. De reden hiervoor is dat het emissiespectrum van overlapt gedeeltelijk die van GFP DsRed. Te omzeilen dit probleem, moet geoptimaliseerd emissie-filters worden gebruikt, dat het minimaliseren van de spectrale overlap tussen DsRed en GFP. Wanneer dergelijke geoptimaliseerde filter sets niet beschikbaar zijn, zou een andere mogelijke oplossing is om een blauw-verschoven fluorescerend eiwit (bijvoorbeeld, BFP of GVB) te gebruiken in plaats van GFP.

Terwijl de muis het netvlies elektroporatie, zowel in vivo en ex vivo, is ontwikkeld en uitvoerig beschreven door Matsuda en Cepko 6-7,9, hebben we recent ontwikkelde een eenvoudige benadering om de activiteit van fotoreceptor-specifieke CRES kwantificeren in geëlektroporeerd muis netvlies 10. Gezien het feit dat de hoeveelheid DNA die in het netvlies geïntroduceerd door elektroporatie kan variëren van experiment om te experimenteren, is het noodzakelijk om onder andere een co-geëlektroporeerd &#39;laden control&#39; in alle experimenten. In dit opzicht is de techniek is zeer vergelijkbaar met de dubbele luciferase assay gebruikt om de promoter activiteit in gekweekte cellen te kwantificeren.
