In het Global Polio Eradication Initiative, laboratoriumdiagnose speelt een cruciale rol door het isoleren en het identificeren van PV uit de ontlasting monsters van acute slappe verlamming (AFP) gevallen. In de World Health Organization (WHO) Global Polio Laboratory Network, PV isolatie en identificatie worden momenteel uitgevoerd met behulp van celkweek systeem en real-time RT-PCR, respectievelijk.
