AFM ist ein hochauflösender (nm-Skala) Imaging-Tool, das mechanisch tastet eine Oberfläche. Es hat die Fähigkeit, Bilder von Zellen und Biomolekülen, in einer flüssigen Umgebung, ohne die Notwendigkeit, chemisch behandeln die Probe. Um dieses Ziel zu erreichen, muss die Probe ausreichend, um die Montagefläche haften, um die Entfernung von Kräften von der Scan-AFM Cantileverspitze ausgeübt zu verhindern. In vielen Fällen hängt die erfolgreiche Bildgebung bei der Immobilisierung der Probe auf der Montagefläche. Immobilisierung von Bakterienzellen auf Gelatine-beschichteten Glimmer ist sehr wahrscheinlich aufgrund von elektrostatischen Wechselwirkungen zwischen der negativ geladenen Bakterien und den positiv geladenen Gelatine. Mehrere Faktoren können mit bakteriellen Immobilisierung, einschließlich der chemischen Bestandteile der Flüssigkeit, in der die Bakterien ausgesetzt, die Inkubationszeit der Bakterien auf die Gelatine beschichtete Glimmer, Oberflächeneigenschaften des Bakterienstammes und dem Medium, in dem die Bakterien sind abgebildet stören. Insgesamt ist die Verwendung von Gelatine-beschichtete Glimmer gefunden allgemein anwendbar zu sein für die Bildgebung mikrobieller Zellen.
