Micro-injectie van lentivirus resulteert in een permanente overdracht van gen-constructen in OSN genomisch DNA. Micro-injectie: Pasgeboren muizen werden gemicroinjecteerd met behulp van een injectiespuit 5μL Hamilton (Hamilton # 7647-01), uitgerust met een 33 gauge naald (Hamilton # 7762-06). Cavia polyklonaal antilichaam tegen VGlut2 (Millipore # AB2251). De ontwikkeling van een nauwkeurige olfactorische circuit is gebaseerd op nauwkeurige projectie van olfactorische sensorische neuron (OSN) axonen naar hun synaptische doelen in de bulbus olfactorius (OB). De moleculaire mechanismen van OSN axongroei en targeting worden niet goed begrepen. Het manipuleren van genexpressie en de daaropvolgende visualiseren van enkele OSN axonen en hun terminal prieel morfologie zijn tot nu toe uitdagend. Om te studeren gen functie op de enkele cel niveau binnen een bepaalde termijn, ontwikkelden we een lentivirale gebaseerde techniek om genexpressie te manipuleren in OSNs in vivo. Lentivirale deeltjes worden geleverd aan OSNs door micro-injectie in het reukepitheel (OE). Expressiecassettes worden dan permanent geïntegreerd in het genoom van getransduceerde OSNs. Groen fluorescerend eiwit expressie herkent besmette OSNs en schetst hun hele morfologie, inclusief de axon terminal prieel. Vanwege de korte doorlooptijd tussen de micro-injectie en verslaggever detectie, kunnen genen functie studies worden geconcentreerd binnen een zeer nauwe periode van ontwikkeling. Met deze methode hebben we ontdekt GFP expressie binnen slechts drie dagen en zo lang als drie maanden na de injectie. We hebben bereikt zowel over-expressie en shRNA gemedieerde knock-down door lentiviraal micro-injectie. Deze methode biedt gedetailleerde morfologie van de OSN cellichamen en axonen op de single cell niveau in vivo, en dus maakt karakterisering van kandidaat-gen olfactorische functie tijdens ontwikkeling.
