Microinjection: souriceaux nouveau-nés ont été microinjectés l&#39;aide d&#39;une seringue Hamilton 5uL (Hamilton # 7647-01) munie d&#39;une aiguille de calibre 33 (Hamilton # 7762-06). Réactifs immunocytochimie: les anticorps primaires: GFP anticorps polyclonal de lapin (Molecular Probes # A-6455), l&#39;anticorps polyclonal dirigé contre le poulet OMP (personnalisé) (Chen et al, 2005.), La Guinée cochon anticorps polyclonal contre VGlut2 (Millipore # AB2251). Anticorps secondaire: Cy2 conjugué d&#39;âne anti-lapin (Jackson Immunolab # 711-225-152), Cy3 conjugué âne anti-poulet (Jackson Immunolab # 703-165-155) et Cy5-conjugués de chèvre anti-Guinée porc (Jackson Immunolab # 106-175-008). Les sections ont été montées sur des lames de verre avec Fluoromount G (Southern Biotech # 0010-01) avec 50ng/mL DAPI chromatine solution de teinture. Développement d&#39;un circuit précis olfactive repose sur la projection précise de olfactifs sensoriels neurone (SVO) axones vers leurs cibles synaptiques dans le bulbe olfactif (OB). Les mécanismes moléculaires de l&#39;OSN axone de croissance et de ciblage ne sont pas bien comprises. L&#39;expression des gènes Manipulation et après la visualisation des axones OSN unique et leur terminal tonnelle morphologie ont été jusqu&#39;à présent difficile. Pour étudier la fonction des gènes au niveau cellulaire unique dans un délai déterminé, nous avons développé une technique basée lentiviraux de manipuler l&#39;expression des gènes dans les ARS in vivo. Particules Lentivirales sont livrés à ARS par microinjection dans l&#39;épithélium olfactif (EO). Des cassettes d&#39;expression sont alors définitivement intégré dans le génome des ARS transduites. Vert l&#39;expression des protéines fluorescentes identifie ARS infectés et expose leur morphologie entier, y compris la tonnelle axone terminal. Grâce au temps de retournement court entre la micro-injection et la détection journaliste, les études de fonction du gène peut être porté dans un délai très étroite du développement. Avec cette méthode, nous avons détecté expression de la GFP dans aussi peu que trois jours et aussi longtemps que trois mois après l&#39;injection. Nous avons atteint deux sur-expression et de médiation shRNA le knock-down par microinjection lentiviraux. Cette méthode fournit des morphologies détaillées des corps cellulaires et des axones OSN au niveau cellule unique in vivo, et permet ainsi la caractérisation de la fonction de gènes candidats au cours du développement olfactif.
