Die Fähigkeit, die Genexpression zu manipulieren ist der Grundstein der modernen experimentellen Embryologie, was zur Aufklärung von mehreren Entwicklungspfaden. Verschiedene leistungsfähige und gut etablierten transgenen Technologien zur Verfügung, um Genexpression in der Maus zu manipulieren, so dass für die Erzeugung sowohl Verlust-und Gain-of-function-Modelle. Allerdings ist die Erzeugung von Maus Transgenik sowohl kosten-und zeitaufwendig. Alternative Methoden der Gen-Manipulation wurden daher häufig gesucht. In utero Elektroporation eine Methode der Gentransfer in die Live-Maus-Embryonen 1,2, dass es uns gelungen, 3,4 angepasst ist. Er basiert weitgehend auf den Erfolg der in ovo Elektroporation Technologien, die häufig in Küken 5 zum Einsatz kommen. Kurz gesagt, ist die DNA in die offenen Kammern des sich entwickelnden Gehirns und die Anwendung eines elektrischen Stromes bewirkt die Bildung von transienten Poren in Zellmembranen injiziert, so dass für die Aufnahme von DNA in die Zelle. In unseren Händen, können Embryonen effizient so früh wie embryonale Tag (E) 11,5 elektroporiert werden, während die Ausrichtung der jüngeren Embryonen eine Ultraschall-geführte Mikroinjektion Protokoll, wie zuvor 6 beschrieben erfordern würde. Umgekehrt ist E15.5 dem neuesten Stand wir leicht elektroporieren kann, aufgrund der Beginn der parietalen und frontalen Knochen Differenzierung, die Mikroinjektion behindert in das Gehirn. Im Gegensatz dazu ist die Netzhaut zugänglich über das Ende der Embryogenese. Embryonen können zu jedem Zeitpunkt während der embryonalen oder frühen postnatalen Phase gesammelt werden. Die Injektion eines Reporter-Konstrukt ermöglicht die Identifizierung von transfizierten Zellen. Bis heute hat in utero Elektroporation wurde am häufigsten für die Analyse der Neokortex Entwicklung 1,2,3,4 verwendet. Neuere Studien haben die embryonalen Netzhaut 7,8,9 und Thalamus 10,11,12 ausgerichtet. Hier präsentieren wir eine in utero Elektroporation Protokoll, das leicht angepasst werden kann, um verschiedene Bereiche des embryonalen ZNS Ziel geändert. Wir beweisen, dass mit Hilfe dieser Technik können wir die embryonalen Telencephalon, Diencephalon und Netzhaut Ziel. Repräsentative Ergebnisse vorgestellt werden, erste, welche die Anwendung dieser Technik, um DNA-Expressionskonstrukte in den Seitenventrikel eingeführt, so dass wir Vorläuferzellen Reifung, Differenzierung und Migration in der embryonalen Telencephalon überwachen.
