1. Specimen collectie <ol><li> Verwijder de vaste palatinale expanders na 4 maanden van intraorale service. Gebruik NOLA Droog veld Systeem om de toestellen te isoleren voordat u ze (figuren 1 en 2). </li><li> Gebruik gesteriliseerde tangen, handschoenen en trays te expanders te verwijderen zonder toevoeging van verontreiniging (figuren 3 en 4). </li><li> Bewaar objecten in Sarstedt 120 ml flacons bij -20 ° C. Biofilm fixatie <ol><li> Schraap biofilm schilfers met een steriele scalpel, of het verzamelen stukken met steriel pincet. </li><li> Voeg ijskoude 4% PFA-oplossing tot monster wordt goed gedekt. </li><li> Incubeer dit mengsel bij +4 ° C (niet invriezen) gedurende 3 tot 12 uur. Uitdroging van vaste monsters <ol><li> Toe te passen 5-30 ul van PFA-vaste monstermateriaal op een objectglaasje. </li><li> Droog bij 46 ° C voor ongeveer 15 minuten of bij kamertemperatuur langer. Ontdooi de lysozym en de formamide. </li><li> Voeg 250 ul van lysozym (1mg/ml) bij kamertemperatuur gedurende 10 minuten, waardoor het gemakkelijker penetratie van de sondes in de cellen door de gedeeltelijke vernietiging van de celwanden. </li><li> Dip glijden in 50%, 80% en 96% (v / v) ethanol gedurende 3 minuten elk. Het uitdrogend effect van de concentratie ethanol serie zal desintegreren van de celmembranen. </li><li> Droog de glaasjes op 46 ° C gedurende 10 minuten. </li></ol> 4. In-situ hybridisatie <ol><li> Bereid 1 mlverse hybridisatie buffer (zie tabel 1 voor de concentraties). Formamide concentraties die in deze studie: 10% (EUBmix), 20% (Bac303, POGI, MUT590) of 45% (LGCmix). </li><li> Ontdooi de oligonucleotide probe-oplossingen. Ontdooide sondes worden bewaard op ijs en beschermd tegen licht. </li><li> Voeg 2 pi van elke probe tot 200 ul van hybridisatie buffer, goed mengen en breng het mengsel op de gedroogde monster op een microscoopglaasje. </li><li> Leg een stukje weefsel papier in een vierkant petrischaal en giet de rest van hybridisatie buffer op het weefsel papier. </li><li> Onmiddellijk horizontaal plaatsen van de dia in de schotel en sluit de schotel. Incubeer in een oven op 46 ° C voor 1-5 uur (90 min volstaan ​​in de meeste gevallen). De schotel fungeert als een vocht kamer het voorkomen van verdamping van hybridisatie-oplossing uit de dia. In het bijzonder kan de verdamping van formamide oorzaak niet-specifieke probe binding aan non-target cellen. </li><li> Bereid 50 ml wasbuffer (zie tabel 2 voor concentrations). Bereid de wasbuffer in een 50 ml buis en verwarm tot 48 ° C in een waterbad. De wasstap wordt uitgevoerd bij 48 ° C. </li><li> Haal de schotel met de dia in de hybridisatie oven. Onmiddellijk wassen uit de hybridisatie buffer met een klein volume van de voorverwarmde wasbuffer, en breng de dia in de resterende wasbuffer. </li><li> Plaats de buis met het wassen buffer en de schuif terug in het waterbad en incubeer bij 48 ° C gedurende 10-15 minuten. </li><li> Verwijder dia uit de tube en dip in de ijskoude DDH 2 O voor 2-3 seconden om de resterende wasbuffer te elimineren. </li><li> De lucht drogen de schuif zo snel mogelijk (het gebruik van perslucht wordt aanbevolen). Sneldrogende zal verminderen sonde dissociatie. </li><li> Gedroogde dia&#39;s kunnen worden opgeslagen in het donker bij -20 ° C gedurende enkele weken zonder een significant verlies van de probe-verleende fluorescentie-signaal. </li></ol> 5. Microscopie <ol><li> Na het FISH en wassen, apply twee druppels antifadent de buurt van de linker-en rechterzijde van een dia (bevroren plakjes moet worden opgewarmd tot kamertemperatuur voorafgaand aan deze stap). </li><li> Plaats een microscoop dekglas op de top en wacht tot de antifadent heeft verspreid over de hele dia. Merk op dat een overmatige hoeveelheid antifadent kan de microscoop beeld vervagen. </li><li> Let op de monsters onder een confocale laser scanning microscoop uitgerust met geschikte filters of lasers. We gebruikten een Leica TCS unit (HCX PL APO/63x; NA 1.2). Gegevens kunnen worden geanalyseerd met software zoals IMARIS of AMIRA. </li><li> Dia&#39;s zijn ingebed in antifadent kan worden opgeslagen bij +4 ° C (niet invriezen) tot 7 dagen voor de probe-verleende fluorescentie begint te dalen. Als alternatief kan de antifadent worden verwijderd met DDH 2 O, en de gedroogde dia&#39;s kunnen worden opgeslagen bij -20 ° C voor een langere periode van tijd. </li></ol> 6. Representatieve resultaten: Schrapen biofilm off vaste orthodontische apparatuur (Figuur 5) geeft geschikt vlokken (Figuur 6), die direct kan worden gehybridiseerd op gecoat glas dia&#39;s voor microscopie. Op deze manier kunnen verschillende groepen orobiome bacteriën geïdentificeerd worden in hun natuurlijke driedimensionale omgeving door tagging bacteriële rRNA met verschillend gelabelde specifieke probes (figuren 7 en 8). In figuur 7 is biofilm gekleurd met EUBmix (groen, alle bacteriën) en LGCmix (geel, Firmicutes). Firmicutes in het groen, als ze waren gekleurd met geel en blauw. In Figuur 8, werd biofilm gekleurd met EUBmix (rood, alle bacteriën), Bac303 (blauw, Bacteroidetes) en POGI (geel, Porphyromonas gingivalis). Porphyromonas gingivalis wordt weergegeven in geelachtig wit, omdat alle drie probes binden aan het DNA en de overlap van kleuren resulteert in een wit signaal. Morfologische verschillen tussen groepen van bacteriën kunnen ook worden geïdentificeerd (figuren 9 en 10). Verschillende vormen van orale bacteriën werden gevisualiseerd in figuur 10, waar coccoid en filamenteuze bacteriën kunnen worden onderscheiden door kleuring met EUBmix (rood). Geen belangenconflicten verklaard.

De beschreven protocol hierin is een zeer bruikbare benadering voor vlekken biofilms verzameld uit harde materialen. Bemonstering en hybridisatie zijn de meest kritische stappen. oral biofilm analysis of palatal expanders by fluorescence in-situ hybridization and confocal laser scanning microscopy

Confocale laser scanning microscopie (CLSM) van natuurlijke heterogene biofilm wordt vergemakkelijkt vandaag door een uitgebreide reeks kleuren van technieken, een van hen is fluorescentie in situ hybridisatie (FISH). 1,2 We hebben een pilot-studie waarin orale biofilm monsters van vaste uitgevoerd orthodontische apparatuur (palatinale expanders) werden gekleurd door FISH, de doelstelling om de drie-dimensionale organisatie van de natuurlijke biofilm en plaque accumulatie te beoordelen. 3,4 FISH creëert een mogelijkheid om cellen vlek in hun eigen biofilm milieu door het gebruik van fluorescent gelabelde 16S rRNA-targeting sondes. 4-7,19 Vergeleken met alternatieve technieken, zoals immunofluorescentie etikettering, dit is een goedkope, nauwkeurige en eenvoudig etikettering techniek om verschillende bacteriële groepen te onderzoeken in gemengde biofilm consortia. 18,20 General probes werden gebruikt die binden aan Eubacteria ( EUB338 + + EUB338II EUB338III; hierna EUBmix), 8-10 Firmicutes (LGC354 AC;. Waren hierna LGCmix), 9,10 en Bacteroidetes (Bac303) 11 Daarnaast zijn specifieke probes binding aan Streptococcus mutans (MUT590) 12,13 en Porphyromonas gingivalis (POGI) 13,14 gebruikt . De extreme hardheid van de betrokken oppervlakte materialen (roestvrij staal en acryl hars) dwong ons om nieuwe manieren van voorbereiding van de biofilm te vinden. Omdat deze oppervlakte-materialen kunnen niet gemakkelijk worden gesneden met een cryotome, werden verschillende bemonsteringsmethoden onderzocht om intact mondeling biofilm te verkrijgen. De meest werkbare van deze aanpak wordt gepresenteerd in deze mededeling. Kleine vlokken van de biofilm-dragende acrylhars werden afgeschraapt met een steriele scalpel, zorg ervoor dat u de biofilm-structuur beschadigen. Tang werden gebruikt om biofilm te verzamelen van de stalen oppervlakken. Eenmaal verzameld, werden de monsters vast en direct geplaatst op polysine gecoat glas dia&#39;s. FISH werd direct uitgevoerd op deze dia&#39;s met de sondes mentioned hierboven. Verschillende FISH protocollen werden gecombineerd en aangepast aan een nieuw protocol, dat was gemakkelijk te hanteren maken. 5,10,14,15 Vervolgens worden de monsters werden geanalyseerd door confocale laser scanning microscopie. Bekende configuraties 3,4,16,17 kan worden gevisualiseerd, waaronder champignon-achtige formaties en clusters van coccoid bacteriën doordrongen door kanalen. Daarnaast werden de bacteriële samenstelling van deze typische biofilm structuren geanalyseerd en 2D-en 3D-beelden gemaakt.
