Geen belangenconflicten verklaard.

In tegenstelling tot veel andere enkele cel technieken kan de time-lapse fluorescentie microscopie hier beschreven methode worden gebruikt om de geschiedenis van een specifieke cel met verwijzing naar zijn voorouders, zijn gedrag, en verdeeldheid gebeurtenissen volgen. De enige beperkingen die we tegenkwamen zijn gerelateerd aan de groeiomstandigheden en de steekproefomvang. Als gevolg van een gesloten omgeving kan medium voorwaarden niet worden gewijzigd tijdens het experiment. Bovendien kan een maximum van vier stammen per experiment efficiënt worden gecontroleerd. Uitgaande van een paar kritische stappen, de single cell analyse hier beschreven methode kan eenvoudig worden toegepast met behulp van een geautomatiseerd microscoop. Het is verstandig om de autofocus instellingen die nodig zijn voor een specifieke bacterie voorafgaand aan het experiment te controleren. Ook moet bij benadering optimale instellingen voor de visualisatie van fluorescentie bepaald worden op voorhand, indien mogelijk. Naar aanleiding een voorbereide time-lijn helpt om al het materiaal klaar voor gebruik in de tijd (pre-opwarming van de microscoop kamer, het programmeren van de microscoop instellingen, het voorbereiden van de glijbaan een uur voordat de cellen in de gewenste groeifase, zie figuur 1) . groei van B. subtilis in TLM en CDM: TLM en CDM zijn chemisch welbepaalde hongersnood media waarin B. subtilis groeit slechts langzaam.
