Dwarslaesie (SCI) schaadt sensorische systemen veroorzaken allodynie 1-8. Om cellulaire en moleculaire oorzaken van allodynie, gevoelige en geldig sensorische tests bij ratten SCI modellen voor nodig is. Echter, tot voor kort had geen enkele testaanpak is gevalideerd voor SCI, zodat gestandaardiseerde methoden niet zijn uitgevoerd in laboratoria. Bovendien kan beschikbare testmethoden niet acuut worden uitgevoerd of wanneer er ernstige motorische beperkingen bestaan, het voorkomen van studies van de ontwikkeling van SCI-geïnduceerde allodynie 3. Hier presenteren we twee gevalideerde sensorische testmethoden waarbij von Frey Hair (VFH) monofilamenten waarvan veranderingen in tactiele zintuiglijke drempels te kwantificeren na SCI 4-5. Een test is de gerenommeerde Up-Down-test, die een hoge gevoeligheid en specificiteit in verschillende SCI ernst wanneer het wordt getest chronisch 5 laat zien. De andere test is een nieuw ontwikkelde dorsale VFH test die acuut kan worden toegepast na SCI wanneer eenllodynia ontwikkelt, voorafgaand aan de motorisch herstel 4-5. Elke VFH monofilament geldt een gekalibreerde kracht bij aanraking met de huid van de achterpoot tot het bochten. In de up-down-methode, worden afgewisseld VFHs van hogere of lagere krachten die op de plantaire L5 dermatoom aan flexor terugtrekking drempels af te bakenen. Achtereenvolgens hogere krachten zijn uitgevoerd tot terugtrekking plaatsvindt, lagere kracht VFHs worden gebruikt tot terugtrekking ophoudt. De voelbare drempelwaarde geeft de vereiste kracht terugtrekking wekken in 50% van de stimuli. Voor de nieuwe test wordt elke VFH toegevoerd aan de dorsale L5 dermatoom van de poot, terwijl de rat wordt ondersteund door de onderzoeker. De VFH stimulatie komt voor in oplopende volgorde van kracht tot ten minste 2 van 3 toepassingen met een bepaalde kracht produceert poot terugtrekken. Tactiele zintuiglijke drempel is de laagste van kracht tot de intrekking 66% van de tijd te lokken. Acclimatisatie, testen en scoring procedures worden beschreven. Afwijkende proeven dat een hertest en typische proeven nodig zijn gedefinieerd. Animal gebruik wzoals goedgekeurd door de Ohio State University Animal Care en gebruik Comite.
