1. Opp / Ned Plantar Von Frey Testing Formålet med testen er å avgjøre taktil følsomhet ved å måle hvor mye kraft som kreves for å lokke fram labben tilbaketrekning fra stimulus. Kommersielt tilgjengelige kalibrerte Von Frey hår monofilamenter mellom utvalgene av 4.56 og 6.10 brukes til å stimulere labben. De VFH testmetoder presenteres her representerer to testing paradigmer som er følsomme og gyldig for bruk i rotte SCI modeller. Ryggmargsskade (SCI) svekker sansesystemer forårsaker allodyni 1-8. Å identifisere cellulære og molekylære årsaker til allodyni, følsom og gyldig sensorisk testing i rotte SCI modeller er nødvendig. Men inntil nylig, hadde ingen enkelt test tilnærming blitt validert for SCI slik at standardiserte metoder ikke har blitt gjennomført på tvers av laboratorier. I tillegg kunne tilgjengelige testmetoder ikke iverksettes akutt eller når alvorlig motorisk svikt eksisterte, hindrer studier av utvikling av SCI-indusert allodyni tre. Her presenterer vi to validerte sensoriske testing metoder ved hjelp av von Frey hår (VFH) monofilamenter som kvantifisere endringer i taktile sansefysiologi etter SCI 4-5. En test er den veletablerte Up-Down test som viser høy sensitivitet og spesifisitet på tvers av ulike SCI alvorlighetsgrader når testet kronisk fem. Den andre testen er en nyutviklet dorsal VFH test som kan brukes akutt etter SCI når enllodynia utvikler, før motor utvinning 4-5. Hver VFH monofilament gjelder en kalibrert kraft når rørt til huden av hind labb før den bøyer. I opp-ned metoden, er vekslende VFHs av høyere eller lavere krefter brukes på plantar L5 dermatome å avgrense flexor tilbaketrekning terskler. Suksessivt høyere krefter brukes til uttak skjer da lavere force VFHs brukes til uttak opphører. Den taktile terskelen reflekterer styrken som kreves for å lokke fram uttak i 50% av stimuli. For den nye testen er hver VFH påføres dorsal L5 dermatome av labben mens rotta støttes av sensor. Den VFH stimulering skjer i stigende rekkefølge av makt til minst 2 av 3 applikasjoner på en gitt kraft produserer pote tilbaketrekning. Taktil sensorisk terskel er den laveste makt for å lokke fram tilbaketrekking 66% av tiden. Akklimatisering, testing og scoring prosedyrer blir beskrevet. Avvikende studier som krever en retest og typisk forsøk er definert. Animal bruk wsom er godkjent av Ohio State University Animal Care og bruk komité.
