rnai mediated gene knockdown and transgenesis by microinjection in the necromenic nematode pristionchus pacificus

Hoewel het meer en meer betaalbaar voor de opkomende modelorganismen volledig gesequenced genoom te verkrijgen, verdere diepgaande genfunctie en expressie analyses van RNA-interferentie en stabiele transgenese blijven beperkt in vele soorten te wijten aan de bijzondere anatomie en moleculaire cellulaire biologie van het organisme. Zo hebben buiten de kroon groep Caenorhabditis Caenorhabditis elegans, dat 3, stabiel overgedragen transgene lijnen in niet-Caenorhabditis soort omvat niet gemeld in dit misleidende phylum (Nematoda), met uitzondering van de Strongyloides stercoralis 4 en Pristionchus pacificus 5. Ter vergemakkelijking van de groeiende rol van P. pacificus in de studie van de ontwikkeling, evolutie en gedrag 6-7, beschrijven we hier de huidige methoden om micro-injectie te gebruiken voor het maken van transgene dieren en gen die door RNAi kloppen. Net als de geslachtsklieren van <em&gt; C. elegans en de meeste andere nematoden, de gonaden van P. pacificus is syncitial en in staat van het opnemen van DNA en RNA in de eicellen bij aflevering door directe micro-injectie. In tegenstelling tot C. elegans echter, stabiele transgene erfenis en somatische uitdrukking in P. pacificus vereist de toevoeging van zelf genomisch DNA gedigereerd met endonucleasen complementair zijn aan de uiteinden van target transgenen en coinjection markers 5. De toevoeging van drager genomisch DNA is gelijk aan de eis voor transgenexpressie in Strongyloides stercoralis 4 en in de geslachtscellen van C. elegans. Het is echter niet duidelijk of de specifieke vereisten van de dieren &#39;eigen genoom DNA is, omdat P. pacificus soma is zeer efficiënt bij zwijgen niet-complexe multi-copy genen of die extrachromosomaal arrays in P. pacificus nodig genomische sequenties voor de goede kinetochoor de montage tijdens de mitose. De ventrale migratie van de twee-armige (didelphic) gonaden in hermafrodieten bemoeilijkt verder de mogelijkheid om zowel gonaden injecteren in de individuele wormen 8. We tonen ook aan het gebruik van micro-injectie om knockdown een dominante mutant (roller, tu92), door het injecteren van double-stranded RNA (dsRNA) in de geslachtsklieren om niet-rolling F 1 nakomelingen te verkrijgen. In tegenstelling tot C. elegans, maar zoals de meeste andere nematoden, P. pacificus PS312 is niet ontvankelijk voor systemische RNAi via voeding en het weken en daarom dsRNA moet worden toegediend door micro-injectie in de syncitial geslachtsklieren. In deze huidige studie, hopen we de micro-injectie proces nodig is om een PPA-EGL-4 promotor te transformeren beschrijven:: GFP fusie verslaggever en knockdown een dominante rol PRL-1 (tu92) mutant in een visueel informatieve protocol.
