In deze video laten we zien hoe de DotScan antilichaam microarray kan gebruikt worden in een eenvoudige, semi-kwantitatieve manier om de oppervlakte-antigeen profielen studie voor celpopulaties van CRC weefsel. De huidige prognose en de classificatie van de CRC is gebaseerd op enscenering systemen die histopathologische en klinische bevindingen te integreren. Echter, in het merendeel van de CRC gevallen cel disfunctie is het resultaat van talrijke mutaties die eiwit-expressie en post-translationele modificatie 1 te wijzigen. Een aantal van de cel-oppervlakte-antigenen, met inbegrip van clustervorming van differentiatie (CD) antigenen, zijn geïdentificeerd als potentiële prognostische of gemetastaseerde biomarkers in CRC. Deze antigenen zijn ideale biomarkers als hun uitdrukking vaak verandert met tumorprogressie of interacties met andere celtypen, zoals tumor-infiltrerende lymfocyten (TIL) en tumor-geassocieerde macrofagen (TAM). Het gebruik van immunohistochemie (IHC) voor kanker sub-classificatie en prognose is goed vastgesteld voor bepaalde typen tumoren 2,3. Er is echter geen enkele &#39;marker&#39; zien prognostische waarde groter is dan klinisch-pathologische staging of algemeen geaccepteerd voor gebruik in de routine pathologie rapportage van alle CRC gevallen. Een meer recente benadering van de prognostische stratificatie van de ziekte fenotypes is gebaseerd op oppervlakte-eiwit profielen met behulp van meerdere &#39;markers&#39;. Terwijl de expressie profilering van tumoren met behulp van proteomics technieken zoals iTRAQ is een krachtig hulpmiddel voor de ontdekking van biomarkers4, is het niet optimaal voor dagelijks gebruik in diagnostische laboratoria en kunnen niet verschillende celtypen te onderscheiden in een gemengde bevolking. Daarnaast worden grote hoeveelheden van het tumorweefsel nodig zijn voor de profilering van gezuiverde plasmamembraan glycoproteïnen van deze methoden. In deze video beschreven we een eenvoudige methode voor oppervlakte-proteomics van levensvatbare cellen van uitgesplitste CRC monsters met behulp van een DotScan CRC antilichaam microarray. De 122-antilichaam microarray bestaat uit een standaard 82-antilichaam regio herkennen van een reeks van lijn-specifieke leukocyten markers, adhesie-moleculen, receptoren en merkers van inflammatie en immuunrespons 5, samen met een satelliet-regio voor de detectie van 40 potentieel prognostische markers voor CRC . Cellen zijn vastgelegd alleen op antilichamen voor die zij uitdragen de bijbehorende antigeen. De cel dichtheid per punt, bepaald door optische scanning, weerspiegelt het aandeel van de cellen die dat antigeen, het niveau van expressie van het antigeen en affiniteit van het antilichaam 6. Voor CRC weefsel of normale darmslijmvlies, optische scans weerspiegelen de immunofenotype van gemengde populaties van cellen. Fluorescentie multiplexing kan vervolgens worden gebruikt om de geselecteerde sub-populaties van cellen van belang gevangen op de array profiel. Bijvoorbeeld, Alexa 647-anti-epitheliale cellen (EpCAM, CD326), is een pan-epitheliale differentiatie antigeen dat gebruikt werd om CRC cellen en ook de epitheliale cellen van de normale darmslijmvlies te detecteren, terwijl de phycoerythrine-anti-CD3, werd gebruikt op te sporen infiltrerende T-cellen 7. De DotScan CRC microarray moet het prototype voor een diagnostisch alternatief voor de anatomisch-based CRC staging systeem.
