Neurale circuits worden georganiseerd in functionele topografische kaarten. Om de complexe schakeling architectuur te visualiseren we een aanpak ontwikkeld om op betrouwbare wijze het etiket van de wereldwijde patronen van meerdere topografische projecties. Het cerebellum is een ideaal model om de ordelijke rangschikking van neurale circuits te bestuderen. Zo heeft de organisatie van de compartimenteel spinocerebellaire bemoste vezels bewezen een onmisbaar systeem voor het bestuderen van bemoste vezels patronen worden. We hebben onlangs aangetoond dat tarwekiem agglutinine (WGA) geconjugeerd aan Alexa 555 en 488 kan worden gebruikt voor het opsporen van spinocerebellaire bemoste vezels projecties in het ontwikkelen en volwassen muizen (Reeber et al.. 2011). We vonden drie belangrijke eigenschappen die de WGA-Alexa tracers wenselijk tools te maken voor de etikettering neurale projecties. De eerste, Alexa fluoroforen zijn intens en de helderheid zorgt voor wholemount imaging direct na tracing. Ten tweede, WGA-Alexa tracers label het gehele traject van het ontwikkelen en volwassen neurale projections. Derde, WGA-Alexa tracers snel vervoerd in zowel de retrograde en anterograde richtingen. Hier beschrijven we in detail hoe de tracers en andere benodigde gereedschappen, hoe u de chirurgie voor spinocerebellaire tracing presteren en de beste manier om de afbeelding getraceerd projecties in drie dimensies te bereiden. Kortom, we bieden een stap-voor-stap tracing protocol dat nuttig zal zijn voor het ontcijferen van de organisatie en de connectiviteit van de functionele kaarten niet alleen in de kleine hersenen, maar ook in de cortex, de hersenstam en het ruggenmerg.
