Eine Methode, um die Position und Orientierung des Submikronpartikel manipulieren destruktiv wäre ein unglaublich nützliches Werkzeug für die biologische Grundlagenforschung werden. Optische Pinzetten, die eine dreidimensionale elektromagnetische Feldgradienten Kräfte auf kleine Partikel ausüben zu nutzen, dies zu erreichen gewünschte Vielseitigkeit und Präzision. 2 Allerdings ist ein großer Nachteil dieses Ansatzes die hohe Strahlungsintensität erforderlich, um die notwendige Kraft zu erreichen, um eine Partikelfalle, die können biologische Proben Schaden 3 Eine Lösung für den Rückhalt und die Sortierung mit geringeren optischen Intensitäten ermöglicht werden optoelektronische Pinzette (OET), aber OET ist außerdem Einschränkungen mit feinen Manipulation von kleinen Partikeln,.. dass DEP-basierte Technologie setzt auch Einschränkung auf dem Grundstück der Lösung 4 , 5 Dieses Video Artikel beschreibt zwei Methoden, die die Intensität der Strahlung für die optische Manipulation von lebenden Zellen benötigt verringern und auch eine Methode zur Orientierung zu kontrollieren. Die erste Methode ist plasmonischen Pinzette, die eine zufällige Gold-Nanopartikel (AuNP) Array zu verwenden als Substrat für die Probe, wie in Abbildung 1 dargestellt. Die AuNP Array wandelt die auftreffenden Photonen in lokalisierte Oberflächenplasmonen (LSP), die von resonanten Dipolmomente, die aus strahlen und erzeugt eine gemusterte Strahlungsfeld mit einer großen Steigung in der Zelle Lösung. Erste Arbeiten zur Oberflächen-Plasmon erweitert Trapping von Righini et al und unsere eigene Modellierung haben gezeigt, den von der Plasmonen Substrat erzeugt reduzieren die anfängliche Intensität erforderlich durch die Stärkung der Gradientenfeld, dass Fallen der Teilchen. 6,7,8 Die Plasmonen Ansatz für feine erlaubt Orientierung Kontrolle von ellipsoidischen Teilchen und Zellen mit niedrigen optischen Intensitäten aufgrund der effizienteren optische Energie Umwandlung in mechanische Energie und einem Dipol-abhängige Strahlungsfeld. Diese Felder sind in Abbildung 2 dargestellt und die geringe Überfüllung Intensitäten sind in den Abbildungen 4 und 5 beschrieben. Die Hauptprobleme bei Plasmonen Pinzetten sind, dass die LSP ist eine beträchtliche Menge an Wärme zu erzeugen und die Trapping ist nur zweidimensional. Diese Wärme erzeugt konvektiven Strömungen und Thermophorese die stark genug sein, um Submikronpartikel aus der Falle zu vertreiben können. 9,10 Der zweite Ansatz, dass wir beschreiben, nutzt periodischen dielektrischen Nanostrukturen einfallendes Licht streuen sehr effizient in Beugung Modi, wie in Abbildung 6 gezeigt, . 11 Idealerweise würde man diese Struktur aus einem dielektrischen Material auf die gleiche Heizung Probleme mit der Plasmonen Pinzette erlebt zu vermeiden, aber in unserem Ansatz eine Aluminium-beschichtete Beugungsgitter ist als eine eindimensionale periodische dielektrische Nanostruktur verwendet. Obwohl es nicht ein Halbleiter, dauerte es nicht erleben eine signifikante Erwärmung und effektiv gefangen kleine Partikel mit geringer Trapping Intensitäten, wie in Abbildung 7 dargestellt. Ausrichtung der Partikel mit dem Gitter Substrat konzeptionell bestätigt die These, dass ein 2-D photonischer Kristall könnte präzise Rotation des nicht-sphärische Partikel mikrometergroßen ermöglichen. 10 Die Effizienz dieser optischen Fallen sind aufgrund der verbesserten Felder durch die Nanostrukturen beschrieben hergestellt erhöht dieser Veröffentlichung.
