synthesis, assembly, and characterization of monolayer protected gold nanoparticle films for protein monolayer electrochemistry

Colloïdaal goud nanodeeltjes beschermd met alkanethiolate liganden genaamd monolaag beschermde goud clusters (MPL) zijn gesynthetiseerd en vervolgens opgenomen in de film assemblages die dienen als platform voor adsorptie eiwit monolayer elektrochemie (PME). PME wordt gebruikt als modelsysteem voor het bestuderen van elektrochemische eigenschappen van de redox-eiwitten door ze aan een adsorptie platform op een aangepaste elektrode, die tevens dienst doet als een redox partner voor electron transfer (ET) reacties. Studies hebben aangetoond dat gouden nanodeeltjes film vergaderingen van deze aard zorgen voor een meer homogene eiwitadsorptie omgeving en het bevorderen van ET zonder afstand afhankelijkheid ten opzichte van de meer traditionele systemen aan te passen met alkaanthiol zelf-geassembleerde monolagen (SAM). 1-3 In deze paper, Mediterrane partnerlanden gefunctionaliseerd met hexanethiolate liganden zijn gesynthetiseerd met behulp van een gemodificeerde Brust reactie 4 en kenmerkt zich met ultraviolet zichtbaar (UV-Vis) spectroscopie, transmissie elektronen microscopie (TEM), en proton (een H) nucleaire magnetische resonantie (NMR). MPC films zijn gemonteerd op SAM gewijzigde goud-elektrode-interfaces met behulp van een &quot;dip cyclus&quot; methode van afwisselende lagen en MPC dithiol koppelen van moleculen. Film groei bij gouden elektrode is elektrochemisch gevolgd door het meten van veranderingen aan de dubbele laag laadstroom van het systeem. Analoge films gemonteerd op silaan gemodificeerde glazen dia&#39;s zorgen voor optische controle van de film de groei en cross-sectionele TEM analyse biedt een schatting laagdikte. Tijdens de film de montage, manipulatie van de MPC-ligand bescherming, alsmede de wisselwerking koppeling mechanisme zorgen voor een netwerk films, die zijn gemakkelijk aan te passen, om te communiceren met redoxeiwit met verschillende adsorptie-mechanisme. Zo kan bijvoorbeeld Pseudomonas aeruginosa azurine (AZ) hydrofoob worden geadsorbeerd aan dithiol gekoppelde films van hexanethiolate mediterrane partnerlanden en cytochroom c (cyt c) kan elektrostatisch worden geïmmobiliseerd op een carbonzuur aangepaste MPC grensvlakken laag. In dit rapport, wij uitsluitend richten op de film protocol voor de AZ-systeem. Onderzoeken waarbij de adsorptie van eiwitten op MPC gemodificeerde synthetische platforms verder kan het begrip van interacties tussen biomoleculen en door de mens gemaakte materialen, en dus steun voor de ontwikkeling van biosensor&#39;s, ET modeling systemen, en synthetische biocompatibele materialen. 5-8
