synthesis, assembly, and characterization of monolayer protected gold nanoparticle films for protein monolayer electrochemistry

Kolloidales Gold-Nanopartikel geschützt mit Alkanthiolat Liganden genannt Monoschicht geschützten Goldcluster (MPL) synthetisiert und anschließend in Film-Baugruppen, die als Adsorptions-Plattformen für Protein-Monoschicht Elektrochemie (PME) dienen. PME ist als Modellsystem für die Untersuchung elektrochemischer Eigenschaften von Redox-Proteine, indem man sie auf eine Adsorption Plattform auf eine modifizierte Elektrode, die auch als Redox-Partner für Elektronentransfer (ET)-Reaktionen eingesetzt. Studien haben gezeigt, dass Gold-Nanopartikel Film Versammlungen dieser Art für eine homogenere Proteinadsorption Umgebung zu schaffen und zu fördern ET ohne Abstandsabhängigkeit im Vergleich zu den traditionellen Systemen mit Alkanthiol selbstorganisierten Monoschichten (SAM) geändert. In diesem Papier und der MPL 3.1 funktionalisiert mit hexanethiolate Liganden synthetisiert werden unter Verwendung eines modifizierten Brust Reaktion 4 und charakterisiert mit UV-sichtbaren (UV-Vis-Spektroskopie), Transmissionselektronenmikroskopie (TEM) und Proton (1 H) Kernspinresonanz (NMR). MPC Filme sind auf SAM modifizierten Goldelektrode Schnittstellen mit einem &quot;dip-Zyklus&quot; Methode der alternierenden MPC Schichten und dithiol Verbindungsmoleküle montiert. Film Wachstum bei Gold-Elektrode elektrochemisch durch die Messung von Veränderungen der Double-Layer-Ladestrom des Systems verfolgt. Analog-Filme auf Silan modifizierte Glasträger montiert ermöglichen optische Überwachung von Film-Wachstum und Querschnitts-TEM-Analyse liefert eine geschätzte Schichtdicke. Während Film Montage, Manipulation der MPC-Ligand-Schutz sowie die interpartikulären Gelenkmechanismus für vernetzte Filme, die leicht anpassbar sind erlaubt, eine Schnittstelle mit Redox-Protein mit verschiedenen Adsorptions-Mechanismus. Zum Beispiel kann Pseudomonas aeruginosa Azurin (AZ) hydrophob zu Dithiol-linked Filme hexanethiolate MPCs und Cytochrom c (Cyt c) können elektrostatisch mit einer Carbonsäure modifizierten MPC Grenzschicht immobilisiert werden adsorbiert werden. In diesem Bericht haben wir auf dem Film-Protokoll für die AZ-System ausschließlich konzentrieren. Untersuchungen, die die Adsorption von Proteinen an MPC modifizierten synthetischen Plattformen könnte weiter das Verständnis der Wechselwirkungen zwischen Biomolekülen und künstliche Materialien, und somit Hilfe zur Entwicklung von Biosensor-Systeme, ET Modellierung von Systemen und synthetischen biokompatiblen Materialien. 5-8
