<p class="jove_content"&gt; Les troubles de la matière blanche du système nerveux central comprend un grand nombre d&#39;étiologies, y compris génétiques, les causes inflammatoires, ischémiques et toxiques<sup&gt; 19,21,22,23,24</sup&gt;. Bien que la sclérose en plaques et vasculopathies sont la principale cause de la maladie de la substance blanche chez les adultes, la leucomalacie périventriculaire associée à la prématurité est la cause la plus fréquente de blessures de la substance blanche dans la population infantile. Afin de mieux délimiter les mécanismes des maladies une étude plus approfondie de la lignée des cellules ofoligodendrocytic, qui sont fortement touchés par l&#39;insulte périnatale, est nécessaire. PEB ont été isolés à partir de la moelle épinière des rongeurs embryonnaire et se sont révélés tripotential dans la nature, donnant lieu à des oligodendrocytes et astrocytes deux populations distinctes, mais ne se différencient en neurones<sup&gt; 2,25,26</sup&gt;. De même, les cellules précurseurs d&#39;oligodendrocytes (OPC) provenant du nerf optique chez le rat et à la fin du cerveau embryonnaire ou postnatale précoce chez le rat ont été démontrées à échéance en oligodendrocytes qui fonctionnent dans les conditions appropriées médias<sup&gt; 27,28</sup&gt;. En outre, fœtales ou adultes dérivées de l&#39;homme OPC ont encore été manipulés en plus matures phénotypes oligodendrocytes<sup&gt; 29,30</sup&gt;.</p&gt;<p class="jove_content"&gt; Cellules GRP ont été isolés à la fois de la moelle épinière de rats et E13.5 E12-13.5 chez la souris. Dans les différences de rat ont été signalés entre les cellules GRP dorsale et ventrale-dérivé dans leur réponse à des conditions qui favorisent la production d&#39;oligodendrocytes, les astrocytes ou avec ventrale dérivés PEB présentant une plus grande propension à se différencier en des phénotypes plus matures<sup&gt; 25,26,28</sup&gt;. C&#39;est peut être dû à la face ventrale de la migration dorsale des cellules GRP naissantes, qui pourraient être moins réceptifs aux signaux de différenciation. Immunopanning permet de la sous-population qui exprime A2B5 à être isolées et recueillies pour le maintien en culture.</p&gt;<p class="jove_content"&gt; Nos souris cultures GRP sont dérivées de tissus de la moelle épinière qui donne au total un groupe hétérogène, la population regroupée de cellules à partir desquelles les cellules GRP sont sélectionnés par des méthodes chimiques (moyennes) et les techniques de base d&#39;anticorps. Bien que nos études ont utilisé la moelle épinière de souris des cellules dérivées GRP, il a plus récemment été rapporté la dérivation de cellules de cerveau de souris progénitrices oligodendrocytes dérivées qui sont également capables de développer en oligodendrocytes matures<sup&gt; 31</sup&gt;. Il s&#39;agit d&#39;un protocole pour la dérivation de la gliales restreints précurseurs (GRP), les cellules de la moelle épinière de fœtus de souris E13. Ces cellules sont les signes précurseurs de la lignée cellulaire oligodendrogliome. Récemment, ces cellules ont été étudiées en tant que source potentielle de thérapies réparatrices dans les maladies de la substance blanche. Leucomalacie périventriculaire (PVL) est la principale cause de non-maladie génétique de la substance blanche dans l&#39;enfance et touche jusqu&#39;à 50% des grands prématurés.
