Dit protocol beschrijft een methode om de enzymatische activiteit van moleculaire chaperonnes te meten in een cel-gebaseerd systeem en de mogelijke effecten van verbindingen met remmende / stimulerende activiteit. Moleculaire chaperones zijn eiwitten die betrokken zijn bij de regulatie van het vouwen van eiwitten 1 en hebben een cruciale rol bij het ​​bevorderen van overleving van de cel op stress-beledigingen als heat shock 2, voedingsstoffen honger en blootstelling aan chemicaliën / gifstoffen 3. Om deze reden chaperones blijken te zijn betrokken bij evenementen zoals de ontwikkeling van tumoren, chemioresistance van kankercellen 4 en 5 neurodegeneratie. Ontwerp van kleine moleculen kunnen remmen of stimuleren van de activiteit van deze enzymen is dan ook een van de meest bestudeerde strategieën voor kankertherapie 7 en neurodegeneratieve aandoeningen 9. De test hier beschreven biedt de mogelijkheid om de hervouwing activiteit van een specifieke moleculaire chaperonne te meten en het effect van de comp studieounds op zijn activiteit. In deze methode het gen van de onderzochte moleculaire chaperone samen getransfecteerd met een expressievector codering voor de vuurvlieg luciferase-gen. Het is al beschreven dat gedenatureerd vuurvlieg luciferase kan worden opnieuw gevouwen door moleculaire chaperonnes 10,11. Als het normaliseren van transfectie controle is een vector die codeert voor de renilla luciferase-gen getransfecteerde. Alle transfecties beschreven in dit protocol zijn uitgevoerd met X-treme Gene 11 (Roche) in HEK-293 cellen. In de eerste stap wordt de eiwitsynthese geremd door het behandelen van de cellen met cycloheximide. Daarna eiwitten ontvouwen wordt veroorzaakt door warmte shock bij 45 ° C gedurende 30 minuten. Na herstel bij 37 ° C, worden eiwitten opnieuw gevouwen in hun actieve conformatie en de activiteit van de vuurvlieg luciferase wordt gebruikt als read-out: hoe meer licht zal worden geproduceerd, de meer eiwitten zal opnieuw kreeg de oorspronkelijke conformatie. Niet-warmte geschokt cellen zijn ingesteld als referentie (100% van hervouwenluciferase).
