MassSQUIRM is een goedkope en kwantitatieve methode voor een uitgebreide analyse van de activiteit van lysine demethylases betrokken bij de mono-en di-methylatie. Onlangs hebben epigenetische regelgevende instanties zijn ontdekt als belangrijke spelers in een groot aantal verschillende ziekten 1-3. Als gevolg hiervan, deze enzymen zijn primaire doelen voor kleine molecule studies en de ontwikkeling van geneesmiddelen 4. Veel epigenetische regelgevende instanties hebben pas sinds kort ontdekt en zijn nog steeds in het proces worden gekwalificeerd. Onder deze enzymen lysine demethylases die methylgroepen uit lysines op histonen en andere eiwitten. Door de nieuwe karakter van deze klasse van enzymen, weinig assays ontwikkeld om de activiteit te bestuderen. Dit is een blokkade om zowel de indeling en hoge doorvoer studie van histon demethylases. Op dit moment zeer weinig demethylase testen bestaan. Degenen die wel bestaan, hebben de neiging om kwalitatief van aard en kan niet tegelijkertijd onderscheid maken tussen de verschillende lysine-methylatie staten (un-, mono-, di-en tri-). Massaspectrometrie wordt vaak gebruikt om demethylase activiteit te bepalen, maar de huidige massaspectrometrische analyses te doenniet in op de vraag of differentieel gemethyleerd peptiden anders ioniseren. Differentiële ionisatie van gemethyleerde peptiden maakt het vergelijken van methylatie staten moeilijk en zeker niet kwantitatief (figuur 1a). Zo beschikbare tests zijn niet geoptimaliseerd voor de uitgebreide analyse van demethylase activiteit. Hier beschrijven een methode MassSQUIRM (massaspectrometrische kwantificering met isotopen reductieve methylering) die is gebaseerd op de reductieve methylering van aminegroepen met gedeutereerde formaldehyde dwingen alle lysines zijn di-gemethyleerde, waardoor zij in wezen dezelfde chemische stof en derhalve de ioniseren zelfde (figuur 1b). Het enige verschil chemische na reductieve methylering waterstof en deuterium, die geen invloed MALDI ionisatie efficiëntie. De MassSQUIRM assay is specifiek voor demethylase reactieprodukten met on-mono-of di-methyl lysines. De test is ook van toepassing op lysine-methyl-geven van de same reactieproducten. Hier wordt een combinatie van reductieve methylering chemie en MALDI massaspectrometrie van de activiteit van LSD1 meten, een lysine demethylase kan verwijderen di-en mono-methyl groepen, een synthetisch peptide substraat 5. Deze test is eenvoudig en lastig om een ​​lab toegang tot een MALDI massaspectrometer lab of via een proteomics faciliteit.
