In dit artikel video beschrijven we een methode voor het bestuderen acute hoornvliesepitheel HSV-1 infectie in een ex vivo model. Dit is een nuttig stap tussen de in vitro en in vivo modellen, omdat daardoor fysiologisch accurate validatie van cel kweek en daarmee beperkt de hoeveelheid dierproeven die moeten worden uitgevoerd. ex vivo organotypic corneal model of acute epithelial herpes simplex virus type i infection

Herpes keratitis is een van de meest ernstige pathologieën geassocieerd met het herpes simplex virus type 1 (HSV-1). Herpes keratitis is momenteel de belangrijkste oorzaak van zowel hoornvlies-afgeleid en infectie-geassocieerde blindheid in de ontwikkelde wereld. Typische presentatie van herpes keratitis omvat infectie van het hoornvliesepitheel en soms de diepere hoornvlies stroma en endotheel, wat leidt tot die vaste hoornvlies pathologieën zoals littekens, dunner, en de dekking 1. Cornea HSV-1 infectie wordt traditioneel bestudeerd in twee soorten experimentele modellen. De in vitro model, waarin gekweekte monolagen van corneale epitheelcellen geïnfecteerd in een petrischaal, biedt eenvoud, hoge reproduceerbaarheid, snelle experimenten en relatief lage kosten. Anderzijds, het in vivo model, waarbij dieren zoals konijnen of muizen rechtstreeks geïnoculeerd in het hoornvlies, biedt een zeer geavanceerde fysiologischesysteem, maar heeft hogere kosten, meer experimenten nodig dierverzorging en een hogere variabiliteit. In deze video artikel we een gedetailleerde demonstratie van een nieuwe ex vivo model van hoornvliesepitheel HSV-1 infectie, die de kracht van beide de in vitro en in vivo modellen combineert. De ex vivo model gebruikt intact cornea organotypically in kweek gehouden en geïnfecteerd met HSV-1. Het gebruik van het ex vivo model maakt zeer fysiologisch gebaseerde conclusies, nog tamelijk goedkoop en vereist tijdsbesteding vergelijkbaar met die van de in vitro model.
