ex vivo mimicry of normal and abnormal human hematopoiesis

Las células madre hematopoyéticas requieren un microambiente único fin de mantener una formación de células sanguíneas, la médula ósea (MO) es un complejo de tres dimensiones (3D) del tejido hematopoyesis en el que se regula por espacialmente organizadas microambientes celulares llamado nichos de 2-4. La organización de los nichos BM es crítica para la función o disfunción de lo normal o maligno BM 5. Por lo tanto una mejor comprensión del microambiente en vivo utilizando una mímica ex vivo nos ayudaría a aclarar los determinantes moleculares, celulares y microambientales de leucemogénesis 6. En la actualidad, las células hematopoyéticas se cultivan in vitro en frascos de cultivo de dos dimensiones (2D) de tejidos o bien placas 7 que requieren ya sea co-cultivo con alogénica o xenogénica las células del estroma o la adición de citoquinas exógenas 8. Estas condiciones son artificiales y difieren de la in vivo </em&gt; microambiente en que carecen de los nichos celulares 3D y exponer las células a las concentraciones de citoquinas anormalmente altos que pueden resultar en la diferenciación y la pérdida de pluripotencia 9,10. Aquí, presentamos una novela de la médula ósea en 3D sistema de cultivo que simula el ambiente in vivo el crecimiento en 3D y es compatible con la hematopoyesis multilinaje en ausencia de factores de crecimiento exógenos. El altamente porosa andamio utilizado en este sistema de poliuretano (PU), facilita la alta densidad crecimiento celular a través de un área de superficie específica mayor que el cultivo en monocapa convencional en 2D 11. Nuestro trabajo ha indicado que este modelo apoyado el crecimiento de sangre de cordón umbilical humano (CB), las células mononucleares (MNC) 12 y primarios células leucémicas en la ausencia de citoquinas exógenas. Esta novela mímica 3D proporciona una plataforma viable para el desarrollo de un modelo humano experimental para estudiar la hematopoyesis y para explorar nuevos tratamientos para la leucemia.
